Description

Insertion of a Stalker element into exon 10 at position 4956 from the start of the longest open reading frame. This generates a stop codon in the vicinity of the insertion site, which is predicted to generate a truncated protein lacking the last 349 amino acid residues (the PHD domains). Transcription is also dramatically lowered.

Insertion of Stalker element.

Detailed Description

e(y)34 mutant embryos exhibit abnormal nuclear division synchronization, with delays in mitosis. This results in morphological disturbances at later stages, even to the degree of the absence of the head.

Homo- or hemizygous e(y)34 mutants exhibit female sterility, decrease in viability, and disturbances in femur development in adults.

Homozygous females and hemizygous males are viable and display a 'bent-leg' phenotype with a penetrance of about 20%.

e(y)34 mutation decreases viability, by 50% in hemizygous males and by 20% in homozygous females. Weak disturbances in the development of the femur, a shortened body and expanded wings are also seen in 15 to 20% of flies. Mutant flies exhibit a repression of heterochromatin silencing.

Mutant flies have a shortened body and spread wings. Males show reduced viability and have defects in femur development.

Flies show slightly reduced viability.

No mutant phenotype in the wing.

Homozygotes display reduced viability.

Stable mutation. Homozygotes have reduced viability and thin bristles. Has no effect on the y+ phenotype.

head and thorax bristles yellow; microchaetae yellow; thin bristles; female sterile; reduced viability.

Genetic Interactions
Statement
Reference

The combination of e(y)34 with Hr46KG00850/+ or Hr46EY05451/+ causes a decrease in viability down to 24 and 8%, respectively.

brm2, mor1 and the combination of Bap170G5986/+ with polybromo33.2/+ act as strong dominant enhancers of the penetrance of the e(y)34/Y bent-leg phenotype in male flies. The combination of e(y)34/Y with brmI21/+ causes lethality.

Hemizygous e(y)34 males display an almost complete suppression of the Pc1 and Pc3 leg transformation phenotype.

Lethal in combination with e(y)11. Viability is rescued by e(y)1+tSa and partially rescued by e(y)11.tSa.

Suppresses the scD1 phenotype slightly in males and strongly in females, including ocellar, post-vertical and scutellar bristles as well as the ANPs. Has no effect on ANP bristle formation caused by y+2MC, sc+2MC and y2#+9, sc2#+9. Has no effect on the Df(1)ase-1, sc5 or sc6 phenotype.

Enhances the cut wing phenotype of ctpN2s and ctn2s, ctpN1 and ctpN30. Acts additively in this enhancement with zv77h. In combination with e(y)34 and zv77h, mod(mdg4)ul enhances the mutant phenotype of ct6, though the gaps in the wing margin are still randomly distributed. No effect on mutant wing phenotype of ct6, ct2s or ct4s.

Heterozygotes with e(y)11 and e(y)21 are lethal. In z1 wzm strain e(y)34 causes a significant reduction of eye pigmentation.

Enhances the y2 phenotype; head and thorax bristles, and hairs are yellow. This enhancer effect is not suppressed by su(Hw)2.

Partially rescued by

e(y)34 is partially rescued by e(y)3ΔPHD

Comments

e(y)3ΔPHD rescues female fertility and viability in the e(y)34 mutant strain and also suppresses the enhancement of the y2 and yinr phenotypes caused by e(y)34.

Comments

Mutation enhances the negative effect of z point mutations, zv77h, z1, zop6 or zop6R1, on w expression, in the presence of certain z alleles e(y)34 becomes dominant. e(y)34 also has a synergistic effect with zv77h on the expression of w (males have orange yellow eyes with red spots). w expression levels of wsp1, wsp2 and wsp4 are moderately reduced, exhibited by a reduced level of eye pigmentation, by e(y)34.
