Description

Point mutation.

Amino acid replacement: K425M. Amino acid replacement: S496A. Amino acid replacement: P454L.

Amino acid replacement: P454L.

Amino acid replacement: K425M. Amino acid replacement: P450L.

Insertion in a presumptive control region.

Carries all the polymorphisms in the z1 sequence as well as the change from Lys to Met; also a C to T mutation causing a proline to leucine change in the middle region of the protein, increasing its hydrophobic nature (Pirrotta, Manet, Hardon, Bickel and Benson, 1987).

Nucleotide change:

C2449828T

Amino acid change:

P450L | z-PA; P450L | z-PB; P450L | z-PC

Reported amino acid change:

P450L

Comment:

Induced on z1 chromosome so also carries a K425M amino acid change. Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change.

Detailed Description

Eye colour: yellow in males, at 25oC.

Eye colour: yellow in males and females.

Do not require two w+ genes for expression unless the z+ gene is present.

Eye colour: zeste in hemizygous males and homozygous females with one or two copies of w+; heterozygous z+/zop6 females wild type if one copy of w+, but zeste if two copies.

Genetic Interactions
Statement

zop6 decreases the eye pigmentation of wP1\loxP.Z.Scer\FRT.Z flies.

Enhances the eye colour phenotype by suppressing the transactivation of the mini-w gene seen when wMcp.y.scs'.1 and ywe.Mcp.2 are combined and when wMcp.scs'.21 and ywe.Mcp.1 are combined leading to lighter eyes.

Partially suppresses the cut wing phenotype of ctpN2s and ctn2s, ctpN1 and ctpN30; acts additively in this suppression with e(y)21. In combination with e(y)21 and zop6, mod(mdg4)ul almost completely suppresses the mutant phenotype of ct6.

Enhancement by e(y)11 and e(y)34 at 18oC; enhancement by e(y)34 shows dominance.

Reverts to weaker alleles after EMS or X rays. Strong antagonist of z+ gene product. Allelic series: zop6 > zop11 > zRN4 > z1 > zRN1 > wild type.

Causes a specific underexpression of w in the eye even when a single, unpaired copy of w is present. Behaves like z+ in promoting transvection effects at Ubx but fails to permit transvection at the dpp locus.

The zop6 mutation causes a new hydrophobic nucleus in the region of the protein preceding the C-terminal domain, resulting in a further stage of multimerization. Alone the S496A and P454L changes have no effect, see z+op6.

Does not alter the effect of mod(mdg4)ul mutation on y2/y+.

Partially inhibits ANP bristle formation caused by y+2MC, sc+2MC and y2#+9, sc2#+9. ANP bristles appear in 70%-90% of flies. Has no effect on the sc phenotype of y2 scD1, y+1MC scD1 or y+3MC scD1 mutants, nor Df(1)ase-1 nor sc6.
