sdETX4 hemizygous males have notched wings.

Hemizygous males have a notched wing phenotype.

Expression of one of sdΔ200.Scer\UAS, sdΔ124-159.Scer\UAS, sdΔ88-159.Scer\UAS, sdΔ160-219.Scer\UAS and sdΔ88-123.Scer\UAS when driven by Scer\GAL4vg.boundary, lead to an enhancement of the wing phenotype seen in sdETX4 mutants. Expression of either sdΔ87.Scer\UAS or sdScer\UAS.cSa driven by Scer\GAL4vg.boundary can either rescue the sdETX4 defective wing phenotype, or can actually make this phenotype more severe, depending on the insertion lines used. The effect of these insertions is also temperature sensitive, with certain insertions suppressing the sdETX4 phenotype at one temperature and enhancing it at another.

sdETX4/Y males display wings with notches at the margin.

Mutants have notches in the wing margin. sd11/sdETX4, sd9/sdETX4 and sd12/sdETX4 flies have a scalloped wing phenotype, with the loss of most of the wing blade tissue and erosion of the wing margin. sd13/sdETX4 flies have a scalloped wing phenotype, with little loss of wing blade tissue but variable erosion of the wing margin (ranging from very conspicuous to loss of only a few margin bristles). sd10/sdETX4 flies have wild-type wings.

Mutant animals have severely notched wings.

Homozygotes show scalloping of the wing at the anterior margin and at the tip. sdSG29.1/sdETX4 flies show slight nicking of the wing, comparable to that seen in sdSG29.1 homozygotes.

Homozygotes have gaps in the wing margin.

Homozygous females and hemizygous males show nicking of the anterior and lateral margins of the wing blade, with males having a more severe phenotype. sdETX4/Df(1)sd72b flies have vestigial wings and the capitellum is reduced. Approximately 85% of sdETX4 flies lack all wing sensilla. Homozygotes and hemizygotes show a reduced response to 1mM sucrose in a proboscis extension test when stimulated on the tarsal or labellar sensilla. Adults also show a reduced response to sucrose, and an enhanced preference and tolerance for salt in a feeding preference assay. The L1 neuron shows a normal response on stimulation of the labellar taste hairs with varying concentrations of sucrose, NaCl or KCl. Larvae show a strongly reduced avoidance of NaCl and KCl in a chemotaxis test. Adults and larvae show a normal response to benzaldehyde and iso-amyl acetate in olfactory assays.

Mutants show a general taste defect. Wings are scalloped.

sdETX4/Y, E2f91/+ males display a severe mutant wing phenotype compared to sdETX4/Y males.

sdETX4/Y males display an enhanced wing phenotype in Df(3R)P-10/+ background.

The addition of sd::vgScer\UAS.sdTEA (driven by Scer\GAL4vg.PM) to sdETX4 animals, almost completely rescues the wing phenotype with only a small amount of wing notching and occasional loss of bristles on the wing margin. The L5 wing vein is also usually incompletely formed. sdETX4, vg1/vg79d5 wings are very small and deformed. The addition of sd::vgScer\UAS.sdTEA (driven by Scer\GAL4vg.PM) significantly rescues the wing defects.

sdETX4 vg79d5 double homozygotes show a significant reduction in wing size and an uplifting of the postscutellar bristles, similar to the phenotype of vg1 homozygotes. Flies heterozygous for vg79d5 in a sdETX4 background have a phenotype almost as strong as sdETX4 vg79d5 double homozygotes. Flies heterozygous for vgnull in a sdETX4 background do not show such a significant reduction in the wing as flies heterozygous for vg79d5 in a sdETX4 or sd1 background. Females expressing vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 in a sdETX4/+ background die in the late pupal stage. They show ectopic wing-like outgrowths at the posterior of the head capsule, with strong deformation of the eye. Males expressing vgScer\UAS.cZa under the control of Scer\GAL4ptc-559.1 in a hemizygous sdETX4 background do not show such extensive wing-like outgrowths and show only slight deformation of the eye. These males can emerge from the pupae, although they die within a few hours.

Expression of Hsap\TDUScer\UAS.cVa under the control of Scer\GAL4ptc-559.1 or Scer\GAL4da.G32 results in a large increase in eye size. This is partially suppressed in a sdETX4 background.

Defects in the wing blade can be rescued by constructs carrying Hsap\TCF13hs.PD.
