Detailed Description

The rough eye phenotype observed in Fmr1sev.PW flies is suppressed in a dcoj3B9/+ background.

Clones of dcoj3B9 are small.

dcoj3B9 clones (generated by the Minute technique) induced in the wing show loss of margin bristles and/or parts of the wing margin. dcoj3B9 clones also show autonomous planar cell polarity wing hair defects. dcoj3B9 Minute clones in the eye show strong planar cell polarity defects.

The eyes of dcoj3B9/dcoar transheterozygotes show mild planar cell polarity defects.

In dcoj3B9 mutant clones in the pupal wing, trichome initiation is delayed and prehairs often emerge from the cell centre instead of the distal edge.

About 2/3 of dcoar/dcoj3B9 flies are arrhythmic, the rest show a period-lengthening phenotype.

Short rhythm mutant.

Homozygous embryos take longer to develop than heterozygous siblings. Foraging motility and touch sensitivity appear normal. The larval lateral neurons (lvLNs) are found slightly more peripherally than in wild type.

Adult-specific expression (using TubGAL80ts) of larkScer\UAS.T:Ivir\HA1 driven by Scer\GAL4P2.4.Pdf significantly lengthens circadian period in dcoj3B9/+ flies; co-expression of larkdsRNA.Scer\UAS.cSb significantly shortens circadian period.

Expression of Nek2dsRNA.Scer\UAS.cUa under the control of Scer\GAL4sd.PU in a dcoj3B9/+ background results in loss of sensory bristles along the wing margin.

The planar polarity defects seen in the eyes of fzhs.sev/+ flies are significantly suppressed in fzhs.sev/+, dcoj3B9/+ flies, although the eyes do not appear wild type.

The planar polarity defects seen in the eye when VangScer\UAS.cWa is expressed under the control of Scer\GAL4hs.2sev in a wild-type background are significantly enhanced in a dcoj3B9/+ background.

Expression of dshScer\UAS.cNa under the control of Scer\GAL4hs.2sev in a dcoj3B9/+ background results in a strong suppression of the loss of photoreceptor cell phenotype seen when the transgene is expressed in a wild-type background.

The ommatidial polarity defects of fz19/fz20 mutants are enhanced in fz19/fz20, dcoj3B9/+ double mutants.

The addition of pers or perT to dcoj3B9/dcoar flies suppresses the arrhythmicity phenotype seen in these flies. The addition of timS1/tim01, tim01/+, or perSLIH does not suppress the dcoar/dcoj3B9 and dcoar/Df(3R)tll-g arrhythmic phenotypes.

Reversion analysis confirms that the P{lacW} insertion is the cause of both the pupal lethality and the aberrant circadian rhythmicity.

After six generations of outcrossing the lethality continued to segragate with the P{lacW} insertion.
