eygM3-12 mutants are headless and have a grooved thorax phenotype.

eyg22-2 in combination with the allele eygM3-12 causes a range of phenotypes in pharate adults: 20% are headless, 35% are eyeless, and 45% have eyes smaller than 400 ommatidia. Furthermore, 100% have a grooved thorax phenotype. The following transgenes cause a dominant enhancement of the grooved thorax and reduced eye phenotype of eyg22-2/eygM3-12 mutants, when expressed under the control of Scer\GAL4eyg-22-2 : eygΔPD.Scer\UAS, eygΔRED.Scer\UAS, eygΔL.ΔRED.Scer\UAS, eygC.Scer\UAS, and eygΔPAI.ΔL.Scer\UAS. eygScer\UAS.T:Hsim\VP16 and eygmPD.Scer\UAS expression enhances the grooved thorax phenotype. Expression of eygmPD.mHD.Scer\UAS or eygScer\UAS.T:Hsim\VP16 causes a headless phenotype with 100% penetrance.

eyg1/eygM3-12 flies have no eyes. eygM3-12 homozygous flies have no head.

eygXE119/eygM3-12 transheterozygotes show a phenotype in which flies have tiny eyes or completely lack eyes altogether. These mutants also have a severe groove on the thorax.

Pharate adults have a headless phenotype - all structures derived from the eye-antennal discs are missing. The prominent remaining structure is the labellum. The fish-trap bristles are also present. In eygM3-12 larvae, no eye-antennal discs can be found. eyg1/eygM3-12 animals exhibit a small eye phenotype.

One copy of eygM3-12 enhances the small eye phenotype seen when Caf1-105NIG.12892R is expressed under the control of Scer\GAL4ey.PH.

Expression of ey::eygGGEGGG.Scer\UAS rescues the eyg22-2/eygM3-12 or ey::eygGGEGEG.Scer\UAS, fails to rescue the mutant phenotype.

DlRevF10, SerRX82 transheterozygotes have normal sized eyes, but eygM3-12; DlRevF10, SerRX82 triple heterozygous flies have small eyes. HE31/+ partially restores eye development in eyg1/eygM3-12 flies, as does NICN.Scer\UAS; Scer\GAL4dpp.blk1 or Su(H)Scer\UAS.cNa; Scer\GAL4dpp.blk1. Head development in eygM3-12 homozygous flies is partially rescued by HE31/+: the resulting flies appear to have a complete head apart from lacking eyes. Head development in these flies is also partially rescued by NICN.Scer\UAS; Scer\GAL4ey.PH : although only 6% of these animals survive to the pharate stage, those that do have heads but usually lack eyes. The eye-antennal discs of eygM3-12/eygM3-12; NICN.Scer\UAS; Scer\GAL4ey.PH larvae have a highly reduced eye field with no photoreceptor differentiation, but an extra eye field with photoreceptor duplication is present dorsal to the endogenous eye field in about a third of cases. In contrast the antennal field of these discs is always present and is sometimes duplicated or triplicated. upd1Scer\UAS.cZa; Scer\GAL4dpp.blk1 completely restores eye development in eyg1/eygM3-12 flies.

The addition of eyScer\UAS.cHa driven by Scer\GAL4ey.PH can partially rescue the eygM3-12/eyg1 mutant eye phenotype.

Complementation and Rescue Data

Expression of eygScer\UAS.lune under the control of Scer\GAL4CD rescues the loss of the eye-antennal disc and headless phenotype of the eygM3-12 mutant. In some cases, duplication of the larval eye-antennal disc and adult antenna occurs. The notum of these animals remains grooved.

Expression of eygScer\UAS.lune under the control of Scer\GAL4EX, Scer\GAL4F1, or Scer\GAL4XS partially rescues the eygM3-12 mutant. In all cases a small head with antennae but without eyes develops. The rescue efficiency of Scer\GAL4XS (76%) and Scer\GAL4EX (60%) are both much higher than Scer\GAL4F1 (7%).

Expression of eygScer\UAS.lune under the combined control of Scer\GAL4EX and Scer\GAL4XS partially rescues the eygM3-12 mutant to a slightly higher percentage (85%) than either driver alone. In this case the same level of rescue is observed, with flies having a small head with antennae but no eyes.

Expression of eygScer\UAS.lune under the transient control of Scer\GAL4CD during stage L2 (expression restricted by Scer\GAL80ts.αTub84B throughout other stages of development) significantly rescues the eygM3-12 eye mutant phenotype. Earlier and later expression are not sufficient to rescue the eye development.

Expression of eygmPD.Scer\UAS, under the control of Scer\GAL4eyg-22-2, partially rescues the eye phenotype of eyg22-2/eygM3-12 mutants, but has a dominant-negative effect on the thorax phenotype. Expression of eygΔHD.Scer\UAS in a eyg22-2/eygM3-12 background leads to partial rescue of the mutant eye and mutant thorax phenotypes. Expression of the eygmHD.Scer\UAS transgene in a eyg22-2/eygM3-12 background leads to the full rescue of eyes in 21% of mutants. The mutant thorax is fully rescued, although there are still many extra macrochaetes at lateral regions. Coexpression of the transgenes eygmHD.Scer\UAS and eygmPD.Scer\UAS in a eyg22-2/eygM3-12 background fully rescues the mutant eye and thorax phenotypes.
