One paternally-derived copy of roX1ex6 reduces the viability of scsisB-1 sisA1/+ females.

The puffing of the male X polytene chromosome that is seen in E(bx)Nurf301-3 mutants is regionally inhibited if they are also mutant for roX1 (carrying roX1ex6); in this case, puffing of a large portion of the distal X, including the region around the roX1 locus, is strongly reduced, while the rest of the chromosome still displays a puffed morphology.

Chromosomal decondensation surrounding autosomal insertions of P{GMroX2} is seen in polytene chromosomes if E(bx)Nurf301-3 is introduced into a roX1- roX2- background (roX1ex6 Df(1)52 P{w+4Δ4.3}) carrying P{GMroX2} insertions.

Only 5% of roX1ex6 Df(1)52 hemizygous males with P{w+4Δ4.3} survive to adulthood, but these animals are abundant and healthy at late third instar. Survival to adulthood is partially rescued (to 11.9%) by msl-1Hsp83.PC; msl-2Hsp83.PK.

The addition of msl-1Hsp83.PC and msl-2Hsp83.PK to roX1ex6 males reduces their viability. roX1ex6, Df(1)52 males have a very low viability, only up to 6.6% of males survive. This loss of viability is partially rescued by adding msl-1Hsp83.PC and msl-2Hsp83.PK, with up to 45% of males surviving.

The majority of roX1ex6; Df(1)52 double homozygous males carrying P{w+4Δ4.3} do not survive to adulthood. This male lethality is almost completely rescued by roX2ΔDHS.Hsp83.

roX1ex6 Df(1)52/Y males carrying P{w+4Δ4.3} generally do not survive to adulthood. This lethality is substantially rescued by an autosomal copy of roX1Hsp83.PM, roX1Δ1.Hsp83, roX1Δ2.Hsp83, roX1Δ3.Hsp83, roX1Δ4.Hsp83, roX1Δ5.Hsp83, roX1Δ6.Hsp83, roX1Δ7.Hsp83, roX1Δ8.Hsp83, roX1Δ9.Hsp83, roX1Δ30.Hsp83, roX1L.Hsp83 or roX1SL.Hsp83. roX1ex6 Df(1)52/Y males carrying P{w+4Δ4.3} generally do not survive to adulthood. This lethality is weakly rescued by an autosomal copy of roX1Δ11.Hsp83. roX1ex6 Df(1)52/Y males carrying P{w+4Δ4.3} generally do not survive to adulthood. This lethality is not substantially rescued by an autosomal copy of roX1a.Hsp83, roX1Hsp83.5', roX1Hsp83.3' or roX1Δ10.Hsp83. roX1ex6 Df(1)52/Y males carrying P{w+4Δ4.3} generally do not survive to adulthood. This lethality is partly rescued by an autosomal copy of roX1ΔO.Hsp83, roX1ΔIR.Hsp83, roX15'S.Hsp83 or roX13'S.Hsp83.

Males carrying roX1ex6,Df(1)52/Y ; P{w+4Δ4.3} exhibit only 5% survival. Df(1)52 deletes roX2 amongst other genes, P{w+4Δ4.3} is a cosmid that contains most of the genes deleted by Df(1)52, but not roX2. Females homozygous for roX1ex6,Df(1)52 and also carrying P{w+4Δ4.3} are viable. The addition of roX2Hsp83.PH to males carrying roX1ex6,Df(1)52/Y ; P{w+4Δ4.3} suppresses the lethality seen in those animals.

Eye colour: the expression of w69C in females carrying w69C in a w- background is unchanged if they are also carrying roX1ex6 Df(1)52 P{w+4Δ4.3}. Eye colour: males carrying w69C in a w- background that are also carrying roX1ex6 Df(1)52 P{w+4Δ4.3} show almost complete derepression of w69C expression in the ventral part of the eye. Eye colour: the expression of wB1-12 in males carrying wB1-12 in a w- background is unchanged if they are also carrying roX1ex6 Df(1)52 P{w+4Δ4.3} (these are escaper males). About 99% of roX1ex6 Df(1)52 P{w+4Δ4.3} males die.
