XX tra1/tra4 pseudomales that are also carrying otuhs.PN and raised at 20-25oC are 2 to 3 times longer than normal. Expression of otuhs.PN partially rescues XX tra1/tra4 pseudomales; 88% of the gonads are 'feminised' and show a developmental progression of gametogenic stages, which can be divided into three sections corresponding to germarial regions 1-3 in normal oogenesis. Section I includes the most apical portion of the pseudomale testis and contains gametogenic stages characteristic of germarial region 1 in ovaries. A group of cells containing spectrosomes is localised at the distal tip. Clusters of cells connected by branched fusomes are located immediately adjacent to the distal cells. The fusomes have disappeared in section II and differentiation of the ring canals continues. In section II, the pseudomale germ cells have differentiated to postgermarial stages. The germ cell clusters contain thick ring canals and the nuclei are large.
Addition of otuhs.PN modestly improves the ovoD1rv23 phenotype. In mutant females homozygous for ovoD1rv23 and with one copy of otuhs.PN, the number of ovary lobes seen with at least one egg chamber increases. However a small number of egg chambers per ovary are seen, with almost all of these displaying a tumorous phenotype, and those without this phenotype have a mixture of tumorous and nurse-like cells. In addition there is some improvement in the degree of oogenic differentiation as the frequency of germ cell clusters with hts positive ring canals or nurse-like cells increases. However chambers remain small and do not progress beyond stage 4. The addition of more than one copy of otuhs.PN have no extra effect. The addition of otuhs.PN to ovoD1rv23/ovoM1, ovoD1rv23/ovoM2 or ovoD1rv23/ovoe8F mothers leads to an increase in the number of eggs produced, though in no case is hatching observed.
One copy can suppress the otu null phenotype at room temperature, allowing egg chamber maturation to late oogenic stages, but typically does not support normal fertility or fecundity.
Complete rescue of both fertility and fecundity were obtained with only the weakest otu allele, otu14. More severe alleles that cause agametic (Df(1)otu-PΔ1 and otu10) or tumorous (otu1, otu3, otu13 and otuPΔ3) phenotypes show substantial morphological suppression but were more variable in their fertility. The same ovarian phenotype is produced with two copies of P{hs-otu.N}.
Partial suppression of the fertility defect does not result from insufficient otu activity.
