    Description

Deletion of 239 base pairs starting from position 999 (by the numbering of cDNA AJ012609). This deletion removes a splice donor site at 1099 bp. The resulting predicted protein, from a transcript using a splice donor site at 907 bp, is lacking amino acids 206-269.

vap2 homozygous adults show significantly decreased body-mass when raised in standard diet, but not in poor diet, as compared to controls. These adults are hypersensitive to acute food deprivation, but not if fed 15% sucrose only.

The maximum lifespan of vap2 homozygous flies is 25 days, compared to 90 days for wild-type controls. Vacuolisation is apparent in brains vap2 homozygous flies by 15 days of age.

vap2/vap2, spri6G1/spri6G1 and vap2/+, spri6G1/spri6G1 double mutants show the same increased resistance to acute food deprivation as spri6G1/spri6G1 single mutants, instead of the hypersensitivity observed in vap2/vap2 single mutants.

A homozygous spri6G1 mutant background significantly suppresses the vap2 neurodegenerative phenotype. Weaker suppression is seen in a spri6G1 heterozygous background.

A heterozygous Rab54 mutant background suppresses the vap2 neurodegenerative phenotype.

A heterozygous Rab52 mutant background significantly suppresses the vap2 neurodegenerative phenotype.

The neurodegenerative phenotype of 18 day old vap2 homozygous flies is dominantly enhanced by styS73 and by rhohs.PSt or EgfrWT.hs under heat shock conditions or Ras85DScer\UAS.cKa; Scer\GAL4Appl.G1a. (Assayed by the percentage of vacuolisation in the optic lobes in 18 day old adults). The neurodegenerative phenotype of 18 day old vap2 homozygous flies is largely suppressed by Egfrf3/+ or drkk02401/+, or by reduction of Egfr levels in adulthood using the temperature sensitive allele Egfrtsla/+. (Assayed by the percentage of vacuolisation in the optic lobes in 18 day old adults). The genetic combination: vap2; Ras85DV12.C40.Scer\UAS; Scer\GAL4Appl.G1a is lethal.

Expression of vapSH2mut32.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav.PU fails to rescue the neurodegeneration phenotype seen in vap2 mutant flies.

Expression of vapScer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav.PU rescues the neurodegeneration phenotype seen in vap2 mutant flies.

Expression of vapSH23mut2.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4elav.PU rescues the neurodegeneration phenotype seen in vap2 mutant flies.
