Mutant embryos derived from homozygous female germline clones have a "lawn of denticles" cuticle phenotype.

pygoS28 mutant embryos that lack maternal and zygotic pygo function display a cuticle denticle lawn that is characteristic of a complete loss of wg signalling.

Somatic clones of pygoS28 homozygous cells in the wing pouch region of third instar wing discs are smaller on average than their wild-type twin spots (P<0.001). Mutant wing pouch clones are associated with increased apoptosis marker expression. When these clones fall in the notum region of this disc there is no difference between mutant clone and twin spot size.

Mutant embryos derived from homozygous female germline clones (lacking both maternal and zygotic pygo function) are short and lack the naked stretches that are normally interspersed between the ventral denticle belts ("lawn of denticles" phenotype). The middle gut constriction is missing. Homozygous clones result in loss of wing tissue, loss of the distal leg, dorsal duplication of the distal leg segments, nicking of the wing and wing margin defects.

The "lawn of denticles" cuticle phenotype seen in pygoS28 embryos is substantially rescued by expression of pan::pygopygoΔPHD-ΔNTCF.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4da.G32; a segmented pattern of denticle bands and naked cuticle is restored. The "lawn of denticles" cuticle phenotype seen in pygoS28 embryos is not rescued by expression of arm::panarmC.ΔNTCF.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4da.G32. The "lawn of denticles" cuticle phenotype seen in pygoS28 embryos is rescued by lgs::pygoΔPHD.HD2.αTub84B.

Overexpression of armScer\UAS.T:Hsap\MYC,T:SV40\nls2, under the regulation of Scer\GAL4da.G32 suppresses the mutant phenotype of pygoS28 embryos which lack both maternal and zygotic pygo function. The degree of suppression varies very little between individual embryos, and is essentially the same as that provided by zygotic pygo expressed from the paternal wild-type allele.

RpS3+; pygoS28 homozygous clones in a RpS32/+ background are 57% the size of RpS3+; pygo+ homozygous clones in a RpS32/+ background; i.e.- pygoS28 clones still have a growth disadvantage even when cell competition is suppressed.

Expression of armS10.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4arm.PS has no effect on the cuticle phenotype of pygoS28 embryos derived from pygoS28 germline clones.
