The mutated residues R4 and E74 fall in the catalytic domain of the wild type protein. E74 lies in the region of the catalytic domain thought to face the protein substrates, not in the region involved in the catalytic activity. dco2 and dco3 share a D401Q polymorphism.

dco3 mutant clones in the eye imaginal disc are overgrown compared to controls.

The overall morphology of dco3 mutants is abnormal, with extensive rippling and loss of normal morphological features including the wing pouch.

dco3/dcot.L39Q transheterozygotes are similar to dco3 mutants.

The rough eye phenotype observed in Fmr1sev.PW flies is suppressed in a dco3/+ background.

dco3 clones in the adult abdomen do not show planar cell polarity defects.

dco3 clones in the wing imaginal disc are larger than their associated wild type sibling clones.

dco3 mutant wing discs collected 36-48 hours after the L2-L3 molt are overgrown.

dco3/dcole88 wing discs exhibit an overgrowth phenotype.

Homozygotes show lethality in larval or early pupal stages, with overgrown discs. Pharate adults generated with dco3/hypomorph combinations show morphogenetic aberrations including duplicated structures. dco3 in combination with dco2 or dco18 : lethality in larval or early pupal stages, with overgrown discs. dco3 in combination with dcole88, dcoi3-193, dcoS053813, dcoS139602, dcoS091510 and dcoP103 : show lethality during larval or early pupal stages with typical dco3 phenotype (swollen tarsi with or without head defects). Discs overgrown. dco3 in combination with dcoS144701 : adult viable with reduced eyes extra vibrissae and expanded wings. dco3 somatic clones in the discs show an autonomous overgrowth phenotype. Cell size is unaffected. Animals with many clones show delayed pupariation.

Hyperplastic overgrowth mutant. Apico-lateral junctional complexes appear normal.

Hyperplastic imaginal disc overgrowth, affecting all discs. Discs implanted into wild type hosts for metamorphosis over grow and differentiate abnormal imaginal structures. dco18/dco3 mutant larvae grow at a normal rate until end of feeding period of last larval instar, when growth slows and then stops: they eventually reach double the weight of wild type pupariating larvae. The ecdysteroid secretory activity of dlg1 and dco ring glands in vitro is 1/6 that of wild type.

Larval life prolonged by up to 14 days. Overgrown larvae show bloating and an increase in water content.

When raised at 25oC the larval period of homozygotes and interallelic heterozygotes is greatly extended and they develop overgrown imaginal discs. Imaginal discs show gap-junctional communication defects: coupling is severely diminished. The single layer epithelium is maintained during overgrowth and they differentiate into the correct but abnormal adult structures.

The lethality and overgrowth phenotypes of dco3 animals are rescued by Scer\GAL4tub.PU-mediated overexpression of wtsScer\UAS.T:Hsap\MYC, resulting in animals that, aside from some mild wing vein phenotypes, are indistinguishable from wild type animals over-expressing wtsScer\UAS.T:Hsap\MYC.
