The mutated residue S181 is strictly conserved in, and specific for, the CKI kinase family. S181 falls in the L-9D loop. This lesion may weaken or prevent the interaction with at least some of the dco substrates, without affecting its catalytic activity.

Hyperplastic imaginal disc overgrowth, affecting all discs. Discs implanted into wild type hosts for metamorphosis over grow and differentiate abnormal imaginal structures. dco18/dco3 mutant larvae grow at a normal rate until end of feeding period of last larval instar, when growth slows and then stops: they eventually reach double the weight of wild type pupariating larvae. The ecdysteroid secretory activity of dlg1 and dco ring glands in vitro is 1/6 that of wild type.
Larval life prolonged by up to 14 days. Overgrown larvae show bloating and an increase in water content.
Hemizygous embryos are non-viable. Some die in the egg case, and have variable head abnormalities. The head is absent and there is a hole in the dorsal larval cuticle in the most severe cases. Homozygous embryos show polyphasic lethality, with most larvae dying around the second-third larval instar transition. A few larvae survive to the third instar, having small imaginal discs with abnormal folds.
When raised at 25oC the larval period of dco18 flies in heteroallelic combinations is greatly extended and they develop overgrown imaginal discs. Imaginal discs show gap-junctional communication defects: coupling is severely diminished. The single layer epithelium is maintained during overgrowth and they differentiate into the correct but abnormal adult structures. Larvae have double mouth hooks, two sets of filzkorper and setal belts and defects around the labrum and dorsal bridge region of the head skeleton.
