Site of nucleotide substitution in mutant inferred by FlyBase curator based on reported amino acid change. A point mutation causing a E509K change in GluRIID, which was not associated with the rhabdomere phenotype, was also identified in this strain.

In pupal and adult Synd661T homozygous mosaic retinas, ommatidia composed of only mutant photoreceptors exhibit C-shaped (fused) peripheral photoreceptor rhabdomeres when compared to controls. In pupal mosaic retinas, all mutant photoreceptors, as well as control photoreceptors neighboring mutant photoreceptors show irregularly rhabdomeres, while most control photoreceptors show the rhabdoemres with the expected round-shape.
In adult Synd661T homozygotes the apical membrane domains of photoreceptors (stalks and rhabdomeres) are disrupted, microvilli of each photoreceptor are not united into a single round rhabdomere, and the catacomb-like membrane architecture is absent, while the basolateral membrane, adherens junctions and cytoplasmic organelles (such as Golgi units and mitochondria) exhibit similar structures when compared to controls. In Synd661T/Df(3R)BSC43 pupae stalk-rhabdomere segregation is also disrupted when compared to controls.
In pupal ommatidia, the heteroallelic combination of Synd661T with Df(3R)BSC43 or Syndmut1 leads to C-shaped peripheral photoreceptor rhabdomeres while Synd661T heterozygosity or the heteroallelic combination of Synd661T with Df(3R)BSC124, Df(3R)BSC141, Df(3R)BSC475, Df(3R)BSC488, Df(3R)BSC516, Df(3R)BSC517, Df(3R)BSC518, Df(3R)BSC680, Df(3R)BSC808, Df(3R)BSC809, Df(3R)BSC818, Df(3R)BSC819, Df(3R)ED10811, Df(3R)ED10820, Df(3R)ED5942, Df(3R)ED6025, Df(3R)ED6027, Df(3R)Exel6184, Df(3R)Exel6185 or Df(3R)Exel6272 does not lead to any alteration in pupal rhabdomare morphology when compared to controls.

In pupal and adult MoePL54 mosaic retinas on a Synd661T/Df(3R)BSC43 background the extent of stalk-rhabdomere segregation defect is not different than Synd661T homozygous mosaic retinas.
In photoreceptor cells of chp2 and Synd661T double homozygous mosaic retinas during pupal stages the stalk-rhabdomere separation is severely and prominently disrupted (even before catacomb-like membrane architecture formation); in adults microvilli show no co-adherence instead spread across the entire apical membrane adult mosaic retina when compared to controls.
Synd661T is rescued by SyndUAS.cUa
Expression of SyndUAS.cUa starting from early pupal development under the control of Scer\GAL4hs.PU rescues the C-shaped rhabdomere phenoype of Synd661T homozygous mosaic retinas while its late expression (starting from mid or late pupal development) under the control of Scer\GAL4hs.PU or Scer\GAL4ninaE.PU fails to rescue the same phenotype.
