Detailed Description

Homozygous clones in the anterior compartment of the wing result in wings which are smaller than normal and which show truncation of vein L5 at the junction with the posterior crossvein.

Rare gbb4/gbbD20 adult escapers have small wings that lack the posterior crossvein and the distal quarter of vein L5.

In gbb4/gbbD20 males raised to adulthood at 18oC and then shifted to 22oC for 7 days, testes contain pn average 1.3 germline stem cells compared to an average of 9.1 in wild-type. Germline stem cells are eliminated completely after a week at 25oC.

In gbb4/gbbD20 females raised to adulthood at 18oC and then shifted to 29oC for 7 days, 60% of germaria lack germ cells. Remaining germaria mostly contain only one germ cell.

myd3PM71/+, gbbD20/+ double heterozygotes show a decrease in flight ability.

The reduced wing size and vein L5 truncation seen in flies carrying homozygous gbbD20 clones in the anterior compartment of the wing are suppressed by scwgbb.PF; the wing is nearly normal in size and vein L5 reaches the wing margin.

The loss of the posterior crossvein and loss of the distal tip of vein L5 which are seen in flies carrying homozygous gbbD20 clones in the posterior compartment of the wing are both partially suppressed by a single copy of scwgbb.PF, resulting in a small fragment of posterior crossvein which is detached from both the L4 and L5 veins and an almost complete L5 vein.

A single copy of scwgbb.PF partially rescues the lethality of gbb4/gbbD20 animals (34% of the expected number of adults are recovered) and the loss of posterior crossvein and truncation of L5 that are seen in gbb4/gbbD20 adult escapers are partially restored. Two copies of scwgbb.PF rescue the posterior crossvein and vein L5 phenotypes to nearly wild type.

scwgbb.PF rescues the lethality of gbbD20/Df(2R)bw-S46 animals; lethality is rescued from the larval/pupal stage to the pharate adult, with rare viable escapers.

bamhs.PO animals cultured at 18oC and heat shocked as late stage pupae (37oC, 4 times for 2 hours each with an 8-12 hour interval) produce adult females with agametic germaria. If these animals also carry the temperature sensitive allelic combination gbb4/gbbD20 and are shifted to 29oC (the non-permissive temperature) for 10 days, no follicle cells remain, although the resulting germaria do contain terminal filament cells.

gbb4/gbbD20 males raised to adulthood at 18oC and then shifted to 22oC for 4 days have an average of 2.4 germline stem cells per testis. This is increased to 4.8 by the presence of DadP1883/+.

gbbD20 dominantly enhances the tkv6 phenotype; tkv6 homozygotes have a normal notum and legs, but the presence of one copy of gbbD20 results in defects in the notum and legs. Double heterozygotes with Df(2L)tkv2, dpps5, shn1 or put135 may show imaginal disc development defects. No interaction with sax5, MadP1 or Med4 is seen in double heterozygous flies.

Dgri\gbb+tFa, Agam\gbb1gbb.PF and Agam\gbb2gbb.PF each completely rescue the lethality of gbbD20/Df(2R)bw-S46 animals.
