Description

A 2.1kb Dp cDNA is expressed from an Hsp70 promoter. The encoded polypeptide is truncated by 8kD relative to endogenous Dp product.

Dpa2 animals partially rescued by Dphs.PD have rough eyes and are female sterile. Nurse cell heterochromatin is also increased. In Dphs.PD egg chambers, unlike wild-type (where nurse cells undergo apoptosis during stages 12 to 13 and have degenerated by stage 14), degeneration of nurse cells is abnormal and occurs late if at all.

Overexpression of both E2fhs.PD and Dphs.PD (using heat shock) in larvae expressing MybΔ.Scer\UAS under the control of Scer\GAL4sd-SG29.1 results in partial rescue of the salivary glands, both with respect to the overall size of the glands and the size of the individual nuclei, and BrdU incorporation can be detected in a number of the salivary gland nuclei with a couple of hours of heat treatment.

Suppresses the DNA replication defects of milouk13910, eIF-4a1006, eIF-4a162-3, eIF-4a1069, eIF-4a1013, bonsaik08322, blwk04409 and alleles of Aats-asp and RpL30.

Co-expression of both E2f1hs.PD and Dphs.PD in starved larvae using heat shock is sufficient to drive most endoreplicating cells into S phase, as assayed by BrdU incorporation. Neuroblasts do not incorporate BrdU in these larvae.

Expression of both Dphs.PD and E2fhs.PD in trolsd larvae using heat shock is able to rescue the proliferation defect seen in trolsd larval brains.

Simultaneous over-expression of E2fhs.PD and Dphs.PD induces DNA replication in epidermal cells of stage 13 or 14 embryos (when these cells are normally arrested in cycle 17). Midgut and salivary gland endocycles are also affected. Over-expression of E2fhs.PD or Dphs.PD alone has no effect. In a CycEAR95 mutant background, no DNA replication is induced in stage 13 or 14 embryos by simultaneous over-expression of E2fhs.PD and Dphs.PD. In a heterozygous CycEAR95 mutant background simultaneous over-expression of E2fhs.PD and Dphs.PD causes induction of S phase in the salivary gland and midgut, but not the epidermis.

Comments

Dphs.PD Partially rescues the lethality seen in Dpa2 mutants.

Daily heat shocks of Dphs.PD in Dpa1/Dpa2 individuals rescues the proliferation defects to normal looking eyes.

Partial rescue of heterozygous lethality at 25oC and full rescue at 37oC.
