egh62d18/egh7 larvae have significantly reduced bouton number and significantly increased bouton size at the neuromuscular junction of muscle 4, compared to controls, with no effect on the number of satellite boutons.

egh7 mutant third instar larvae exhibit enlargement of peripheral nerves attached immune cells. This is most pronounced in the peripheral nerves that project from the most caudal segment (A9) of the ventral nerve cord. The immune cells are identified as plasmatocytes, that are normally circulating in the hemolymph. In severe cases, large plasmatocyte aggregates completely enfold the nerves in the mutants. There is no obvious correlation between nerve diameter and the degree of plasmatocyte accumulation in the mutants. Neither nerve growth nor plasmatocyte accumulation is observed in wild-type larvae.

eghcm/egh7 females are insensitive to injection with sex peptide; oviposition is not induced in these females and mating is not suppressed.

Remating in eghcm and eghcm/egh7 females is about 48% and 24% compared to mating of virgin eghcm and eghcm/egh7 females, which is a higher amount than in control females. Sex appeal of mated eghcm/egh7 females to males is reduced to the same extent as control mated females.

egh7 clones in the VNC Ap neurons do not extend neuronal projections to the anterior part of the central brain and fail to form the elaborate arborizations that are observed in wild-type flies.

In larval brains of eghcm/egh7 mutants, Ap neurons projecting from the VNC to the CNS show pathfinding errors and fail to reach the target area. In many cases, these axons defasciculate and turn backward. Mostly, these axons stop extending toward the central brain but rarely defasciculate.

In the brains of egh7 third instar larvae, some of the axons of R-cells 1-6 fail to terminate in the lamina and project into the medulla, causing thick axon bundles in this structure. R1-R6 axons that do terminate in the lamina are disorganized and form abnormal patches in the lamina plexus. In egh7 mutants, the boundary region that exists in wild type between the lobula and lamina is no longer apparent and the sheath-like glial processes that usually delimit this boundary are severely disrupted or missing altogether. This allows lobula distal neurons to cross into the base of the developing lamina, disrupt the pattern of lamina glia and result in the perturbed R1-R6 axonal projections. Clonal analysis shows that the egh7 is required in cells of the lobula complex primordium, as egh7 mutant clones in this structure are associated the same perturbed R1-R6 axonal projection phenotype as egh7 flies. However, egh7 clones in the eye, glial cells, lamina precursors, or lamina neurons do not cause this phenotype.

Females with germline clones lay eggs with fused dorsal appendages and the embryos have an expanded nervous system. Only a few eggs are laid because early oogenesis is blocked due to the development of egg chambers containing multiple oocyte-nurse cell complexes. Germ cell divisions in the egg chambers are not regulated. Follicle cells at the posterior of egg chamber become mesenchymal-like indicating they have not adopted a border cell fate. Stalk cells are determined correctly but are usually disorganised compared to wild type.

In egh7/Y; sli2/+ and egh7/Y; slik04807b/+ mutants, the R-cell axonal projection pattern is defective like in egh7 single mutants. When egh7 is heterozygous in these double mutants, flies appear wild type. Expression of robodsRNA.Scer\UAS, under the control of Scer\GAL4elav-C155, does not alter the R-cell phenotype of egh7 mutants.

Most eghcm/egh7 larval brains show a complete rescue of the ventral nerve cord defect when eghScer\UAS.T:Ivir\HA1 in expressed under the control of Scer\GAL4ap-md544.

Expression of eghScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tsh-md621 rescues the viability of egh7 flies but these flies are short lived. Scer\GAL4ap-md544>eghScer\UAS.T:Ivir\HA1 expression does not rescue egh7 viability.

Expression of eghScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4tsh-md621 rescues the receptivity response of eghcm/egh7 females to sex peptide completely. This response is partially rescued when eghScer\UAS.T:Ivir\HA1 is expressed under the control of either Scer\GAL4tub or Scer\GAL4ap-md544, but is not rescued when Scer\GAL4elav.PLu is used to drive expression. Expression of eghScer\UAS.cFa under the control of Scer\GAL4elav-C155 rescues the R-cell projection phenotype in 80% of mutant egh7 brain hemispheres tested.

Lethality and fertility phenotypes are rescued by egh+t7 and eghhs.PG.

Lethality is rescued by egh+t7, rescued adults are fertile.
