UAS regulates expression of a 't-peptide' comprising (from N to C terminus): a trypsin secretory signal sequence for targeting to the secretory pathway, a mature cleaved Pdf peptide sequence, a medium length hydrophilic linker sequence of 14 amino acids with an embedded Tag:MYC epitope tag, and a GPI targeting sequence from lynx1 protoxin.

Expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4GMR18H11 significantly suppresses sleep late at night. This effect is blocked if the flies are also carrying Scer\GAL80cry.PS.

Flies expressing PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4nSyb.PU have free-running circadian rhythms with a strength and period indistinguishable from controls.

Males expressing PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4desat1.PB show a significant increase in the levels of (z)-7-Tricosene and (z)-7-Pentacosene at all circadian time points compared to wild type (measured on the sixth day of constant darkness).

Scer\GAL4tim.Scer\UAS-mediated expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 induces complex free-running circadian locomotor rhythms in flies in constant darkness.

Scer\GAL4Mz1366- or Scer\GAL4Mz1525-mediated expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 fails to induce free-running circadian locomotor rhythms in flies in constant darkness.

Scer\GAL4tim.Scer\UAS-mediated expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 in the presence of Scer\GAL80Pdf.PS (i.e. expression in all clock neurons other than the Pdf-expressing LN[[V]]s) induces complex free-running circadian locomotor rhythms in flies in constant darkness.

Scer\GAL4cry.PZ (P{cry-GAL4.Z}24)-mediated expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 induces complex free-running circadian locomotor rhythms, with a short period of 22-23 hr and a long period of 25-26 hr, in flies in constant darkness. In contrast, Scer\GAL4cry.PZ (P{cry-GAL4.Z}16)-mediated expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 induces only a modest degree of complex rhythmicity and rather induced dramatic period shortening of 3-4 hr.

Expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under either the combined control of Scer\GAL4Clk.int1-3 and Scer\GAL80Pdf.PS, or the control of either Scer\GAL4Pdf.PU, Scer\GAL4Clk.4.1M or Scer\GAL4Clk.4.5F, does not rescue the lower night-onset preferred temperature decrease observed in Dh3151 homozygous mutant adults.

The suppression of total nighttime sleep caused by expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4GMR18H11 is abolished in a Dh31KG09001 homozygous background, although there remains a small, statistically significant decrease in sleep during the second half of the night.

Pdfr5304 does not suppress the increase in the levels of (z)-7-Tricosene and (z)-7-Pentacosene compared to wild type seen in males expressing PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4desat1.PB (measured on the sixth day of constant darkness).

Co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 rescues rhythmic locomotor behaviour under constant darkness conditions in 67.5% of flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The locomotor activity defects seen under 12 hour light/12 hour dark conditions in flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) are not suppressed by co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1.

Co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 rescues rhythmic locomotor behaviour under constant darkness conditions in 41.0% of flies expressing gwHMS00105 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency).

Pdf01 flies that also express PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4tim.Scer\UAS exhibit little arrhythmicity in constant darkness (unlike Pdf01 only flies), with only ~10% of flies arrhythmic and most exhibiting instead a complex free-running rhythm (which is still abnormal compared to wild type).
