The expression of gwKK101472 under the control of Scer\GAL4c564 has no obvious affect on the inflammatory response to Gram-positive bacterial infection or under basal conditions, as compared to controls.

Flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency) are arrhythmic for locomotor activity under constant darkness conditions.

Circadian behaviour under 12 hour light/12 hour dark conditions is altered in flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) The morning peak of activity is severely blunted, although a robust evening peak is still present. The phase of the evening peak of activity is advanced compared to control flies.

Only 25% of flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) and in the presence of Scer\GAL80Pdf.PS (to further restrict Scer\GAL4 expression) are rhythmic for locomotor activity under constant darkness conditions. Under 12 hour light/12 hour dark conditions morning anticipation is decreased and the phase of the evening peak of activity is advanced compared to controls.

Flies expressing gwKK101472 under the control of Scer\GAL4P2.4.Pdf show normal circadian locomotor behaviour under both 12 hour light/12 hour dark and constant darkness conditions.

Co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 rescues rhythmic locomotor behaviour under constant darkness conditions in 67.5% of flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency). The locomotor activity defects seen under 12 hour light/12 hour dark conditions in flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa) are not suppressed by co-expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1.

Co-expression of PdfScer\UAS.SCR.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 does not rescue rhythmic locomotor behaviour under constant darkness conditions in flies expressing gwKK101472 under the control of Scer\GAL4tim.PE (in the presence of Dcr-2Scer\UAS.cDa).
