Detailed Description

Dh31KG09001/Dh31KG09001 and Dh31KG09001/Df(2L)Exel7038 flies show significantly more nighttime sleep than controls (the amount of daytime sleep is unaffected). The increase in nighttime sleep occurs in the second half of the night in the mutant flies. The nighttime sleep is more consolidated than that of controls, with the mutant flies showing fewer sleep bouts of longer duration. The defects are more severe in the hemizygous Dh31KG09001/Df(2L)Exel7038 flies than the homozygous Dh31KG09001 flies.

The sleep-suppressing effect of carbamazepine is strongly attenuated in homozygous flies compared to controls.

Dh31KG09001/Dh31KG09001 and Dh31KG09001/Df(2L)Exel7038 flies have free-running circadian rhythms with a strength and period indistinguishable from controls.

The suppression of total nighttime sleep caused by expression of PdfScer\UAS.ML.T:SS-trypsin.T:Hsap\MYC.T:GPI-lynx1 under the control of Scer\GAL4GMR18H11 is abolished in a Dh31KG09001 homozygous background, although there remains a small, statistically significant decrease in sleep during the second half of the night.

Comments

Expression of Dh31Scer\UAS.cKa under the control of either Scer\GAL4GMR18H11 or Scer\GAL4GMR20A02 rescues waking during the 3 hours before dawn in homozygous Dh31KG09001 flies.
