Description

FLP-mediated recombination between the FRT sites of PBac{WH}f00735 and PBac{WH}Nlg1f00713 generates an approximately 29kb deletion within Nlg1 that removes the transcription start site and most of the extracellular domain of the protein.

Nlg1Δ46 homozygotes are viable. Nlg1Δ46 leads to a significant changes in the third instar larval neuromuscular junction compared to controls: a decrease in the number, but not in the area, of synaptic boutons; a significant increase in the active zone density; a significant increase in the total postsynaptic density length; a significant increase in the ruffle density at the synapse; and significant decreases in the subsynaptic reticulum length and density.

Nlg1Δ46 mutants exhibit smaller, weaker synapses than in wild-type.

Nlg1Δ46/Df(3R)ED5223 mutants display a reduction in synaptic boutons compared to controls.

Nlg1Δ46 homozygotes and Nlg1Δ46/Df(3R)ED5223 transheterozygotes exhibit a significant reduction in the amplitude of EJCs evoked by nerve shock, compared to wild-type. Amplitude distributions of miniature EJCs (mEJCs) are abnormal in Nlg1Δ46 mutants, displaying a significantly broader distribution, compared to wild-type. Despite this change in distribution, the mean amplitude of mEJCs remains as in wild-type. Therefore, the primary affect of Nlg1Δ46 is to reduce the quantal content, the number of vesicles released by action potential, by more than 60%.

Ca[2+] sensitivity of vesicle fusion is unaltered in Nlg1Δ46 mutants. The primary difference between mutant and wild-type is the size of synaptic current at maximal release.

Nlg1Δ46, Nrx-1273 double homozygotes show third instar larval neuromuscular junction defects that are similar to those observed in either single mutant, although there are ocasional giant boutons that are laterally elongated with multiple active zones and reduced subsynaptic reticulum; Nrx-1273, Nlg1Δ46 double heterozygotes also present a significant decrease in the number of synaptic boutons at the third instar larval neuromuscular junction, as compared to single heterozygous and wild-type controls.

Nlg1Δ46 homozygosity does not suppress or enhance the third instar larval neuromuscular junction defects displayed by witB11/witA12 transheterozygotes. Nlg1Δ46 homozygosity also does not suppress or enhance the decreased number of synaptic boutons at the third instar larval neuromuscular of Mad12/Madk00237 transheterozygotes.
