In the asrij9/asrij9 third instar larval lymph gland, mitochondria show increased branch length, footprint (content), number of branches, number of junctions and aggregation. The lymph gland shows an increase in crystal cells in the primary lobe.

Hemocytes from asrij9 homozygous mutant larvae do not display any obvious defects in their phagocytic ability.

asrij9 mutants show premature differentiation to P1+ plasmatocytes.

Homozygous third instar larvae have a higher number of secondary lymph gland lobes than normal which expand into posterior segments up to A4 or A5 (excess secondary lobes are not seen in mutant embryos or second instar larvae). The mutant ectopic lobes are asymmetric and extend up to three quarters of the length of the dorsal vessel.

The number of mitotic foci in the primary lymph gland lobe of mutant third instar larvae is normal, but there is a dramatic increase in the number of mitotic foci in the secondary lymph gland lobes at this stage.

The primary lymph gland lobe of mutant third instar larvae show premature loss of the medullary zone and hemocyte differentiation compared to controls.

The total number of circulating hemocytes is significantly increased in mutant third instar larvae compared to wild type.

Transgenic flies that express Scer\GAL4e33C>Mmus\Cd8aScer\UAS.T:Avic\GFP in asrij9 mutants show complete suppression of premature plasmatocyte differentiation.

Comments

Expression of asrijScer\UAS.cKa under the control of either Scer\GAL4e33C or Scer\GAL4Hml.PG restores lymph gland lobe number and pericardial cell arrangement to a near wild-type pattern in asrij9 third instar larvae.
