Expression of mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4He.PZ results in hemocytes exhibiting abnormally enlarged endolysosomal compartments.

Expression of mtmdsRNA.cVa.Scer\UAS under the control of either Scer\GAL4how-24B or Scer\GAL4Mef2.PR results in a decrease in the number of both dorsal temporary internal oblique muscles (IOMs) and ventral persistant larval muscles (PLMs) in the abdomen of pharate adults. The IOMs that are present are frequently detached. Central myofibrils are misaligned or absent around a normal number of centrally-displaced nuclei (distance of nuclei from midline is increased) in the IOMs of pharate adults expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Mef2.PR. The nuclei are otherwise normal in size and morphology, mitochondrial integrity is normal and the peripheral myofibrils appear normal. Transverse (T-) tubules are disrupted.

IOMs expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Mef2.PR are properly maintained during early pupal stages, when other larval muscles undergo developmentally regulated cell death (2 days after puparium formation, APF) and subsequently undergo normal myofiber thinning (at 3 days APF) and rethickening (at 4 days APF), but IOM muscle detachment is seen during remodeling in late pupal stages (4 days APF).

Visceral muscles that normally migrate to ensheath the testis are present but disrupted in adults expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4how-24B.

Primary hemocytes dissected from wild-type larvae undergo remodelling from a round to a spread cell shape, having F-actin rich radial protrusions. However, primary hemocytes derived larvae expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Pxn.PS lack these protrusions.

Third instar larvae expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Pxn.PS have a normal number of bled hemocytes per larva and have a normal number of sessile hemocytes per segment, although the sessile hemocytes have an abnormally clumped distribution compared to controls.

Third instar larvae expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Pxn.PS have a reduced number of hemocytes recruited to a puncture wound site 6 hours after wounding compared to control larvae.

Primary hemocytes derived from larvae expressing mtmdsRNA.cVa.Scer\UAS under the control of Scer\GAL4Cg.PA have enlarged endolysosomes.
