Pdfr5304 hemizygotes show severe and nearly sully penetrant circadian arrhythmicity and a significantly shorter circadian period.

Pdfr5304/Y mutant adults have significantly reduced strength of circadian locomotor rhythms under constant darkness, compared to heterozygous controls.

Pdfr5304 homozygous mutant adults, reared under 12:12 light:dark zeitgeber cycle, present a lower night-onset preferred temperature decrease (between zeitgeber times 10-12 and 13-15) but similar day-time preferred temperature increase, as compared to controls.

In contrast to wild-type, Pdfr5304 mutants fail to exhibit any light-dependent temperature preference.

In constant darkness, almost a quarter of Pdfr5304/Pdfr5304 flies are arrhythmic and almost half have weak rhythms.

Mutant flies show severely reduced morning anticipation and the phase of the evening peak of activity is advanced compared to controls when circadian locomotor activity is analysed under 12 hours light/12 hour dark conditions. The flies are mostly arrhythmic under constant darkness conditions.

The E pacemaker neurons in the brain of mutant flies show no response to Pdf, in contrast to wild type.

Pdfr5304 mutants exhibit daytime temperature preference rhythms similar to those of wild-type flies during LD.

Mutant flies maintain a morning peak of circadian locomotor activity under 12 hour light:12 hour dark conditions, but show an advanced evening peak of activity compared to wild-type flies. 59.3% of the mutant flies are arrhythmic in constant darkness conditions (measured during days 3-9).

pdfr5304 mutants exhibit phase-advanced evening anticipation.

Behaviour rhythms of pdfr5304 mutants damp in DD and are accompanied by a shorter period.

Under light:dark (LD) conditions, mutant flies show increases in sleep during the late night. At the time when wild-type flies are waking up in advance of lights-on, the mutant flies fail to wake up.

During the first day of constant darkness (DD) conditions, mutant flies show a significant increase in total sleep during the subjective day compared to control flies.

Homozygotes display strongly reduced morning anticipation and phase-advanced evening anticipation under conditions of light-dark entrainment and on the first day of constant darkness.

pdfrEY11181-GAL4/pdfr5304 flies display poor rhythmicity in constant dark conditions.

pdfr5304 is viable and does not show any atypical morphological phenotype.

pdfr5304 flies appear to constantly prefer a temperature of 23.5oC, regardless of whether it is day or night. This is a little colder than the temperature preferred by wild-type flies.

pdfr5304 mutants show reduced behavioural activity in the morning during LD cycles and during the first day of the DD cycle; the average activity during the first wo hours after 'light on' is 10% of the total daily activity in LD. More than 60% of pdfr5304 flies show less than 10% of their total daily activity during the first two hours in the morning. pdfr5304 flies do not demonstrate any gradual increase of actvity before lights are turned on, indicating that these mutants have a deficit in morning anticipation.

pdfr5304 mutants display an abnormality in evening behavioural activity. They begin evening activity earlier than wild-type flies and the activity continues until lights are turned off in LD cycles. The highest evening activities are observed at ZT9.6, compared to ZT11.0 in control flies, suggesting that evening anticipation is advanced in pdfr5304 mutants.

pdfr3369 mutant flies exhibit abnormal circadian rhythmic behaviour under DD conditions. They show residual circadian rhythmic behaviour in the first few days, but they gradually lose the rhythmic activity as the DD condition continues. The rhythmic power of the pdfr3369 mutant is 69 in DD, compared to 121 in control flies. pdfr3369 mutants demonstrate a shortened rhythmic period of 22.7 hours, compared to 23.3 hours in wild-type. The rhythmicity during D is observed in only 10-15% of individual flies tested in the pdfr3369 genotype.

Expression of PdfrScer\UAS.cMa under the combined control of Scer\GAL4Clk.int1-3 and Scer\GAL80Pdf.PS, but not under the control of either Scer\GAL4Clk.4.5F or Scer\GAL4Pdf.PU, rescues the lower night-onset preferred temperature decrease observed in Pdfr5304 homozygous mutant adults; expression under the combined control of Scer\GAL4Clk.int1-3 and Scer\GAL80Pdf.PS also significantly exacerbates the day-time preferred temperature increase observed in Pdfr5304 homozygous mutant adults.

Expression of PdfrScer\UAS.cMa in DN1[[p]]s under the control of Scer\GAL4Clk.4.5F restores normal light-dependent temperature preference in Pdfr5304 mutants.

PdfrT:Hsap\MYC rescues both the advanced evening peak of locomotor activity which is seen in Pdfr5304 flies under light-dark conditions and the arrhythmicity of Pdfr5304 flies under constant darkness conditions.

In contrast, morning anticipation is resuced by both Scer\GAL4Clk.4.1M and Scer\GAL4Clk.4.5F expression of pdfrScer\UAS.cZa.

Behaviour rhythms of pdfr5304 mutants are rescued by Scer\GAL4Clk.4.1M-driven expression of pdfrScer\UAS.cZa. There is significant improvements of rhythm amplitude with Scer\GAL4Clk.4.1M-driven expression of pdfrScer\UAS.cZa but not with Scer\GAL4Clk.4.5F-driven expression.

Expression of pdfrScer\UAS.cMa using Scer\GAL4Clk.8.0 in a pdfr5304 background rescues the defects in morning anticipation and the timing of light-dark entrained evening behaviour.
