The expression of paraGD3392 under the control of Scer\GAL4elav.PU alone leads to a significant decrease in climbing distance in negative geotaxis assays as compared to controls, whereas expression under the combined control of Scer\GAL4elav.PU and Dicer-2 transgene (to enhance RNAi efficiency), results in lethality before adulthood.

results in defects at the neuromuscular junction of third instar larval abdominal muscles 6 and 7: expression under the control of Scer\GAL4elav.PU alone results in a strong increase in the number of synaptic boutons of >=5micron diameter, despite a small but significant decrease in the total number of boutons, a strong increase in the number of 'ghost' boutons and a small but significant decrease in the number of branches, as compared to controls; expression under the control of Scer\GAL4elav.PU results in a significant decrease in the frequency, but not amplitude, of miniature excitatory junction potentials and results in a significant increase in the latency period to stimuli, but not in the amplitude of corresponding excitatory junction potentials, as compared to controls.

Expressing paraGD3392 under the control of Scer\GAL4GMR.PU leads to a subtle rough eye phenotype.

Individuals expressing paraGD3392 under the control of Scer\GAL4elav.PU exhibit delayed eclosion

Expression of paraGD3392 under the control of Scer\GAL4elav.PLu decreases the penetrance of the bang sensitive phenotype seen in parabss1/Y males ((from 100% to 65%) and parabss1/+ females (from 87% to 1.6%).

Expression of two copies of paraGD3392 under the control of Scer\GAL4elav-C155 results in complete lethality.

Expression of two copies of paraGD3392 under the control of Scer\GAL4insc-Mz1407 results in flies being reversibly paralysed at 38[o]C.

Adults expressing paraGD3392 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Reference

The co-expression of paraGD3392 and Cnx99AKK108301 under the control of Scer\GAL4elav.PU alone leads to male lethality and enhances the locomotor defects in negative geotaxis assays exhibited by females expressing paraGD3392 alone.

At the neuromuscular junction of third instar larval abdominal muscles 6 and 7, the co-expression of paraGD3392 and Cnx99AKK108301 under the control of Scer\GAL4elav.PU partially suppresses the increased total number of boutons induced by the expression of Cnx99AKK108301 alone, and suppresses the increased 'ghost' bouton number, but not the increase in the number of synaptic boutons of >=5micron diameter, the decrease in the total number of boutons or the decrease in the number of branches, induced by the expression of paraGD3392 alone; the co-expression of paraGD3392 and Cnx99AKK108301 under the control of Scer\GAL4elav.PU does not significantly enhance the increased latency period to stimuli induced by the expression of either Cnx99AKK108301 alone or paraGD3392 alone, but partially suppresses the decreased miniature excitatory junction potentials induced by the expression of either Cnx99AKK108301 alone.

Crei\ChR2Scer\UAS.cHa.T:Avic\GFP-EYFP larvae expressing paraGD3392 under the control of Scer\GAL4ppk.1.9 exhibit a reduction in nocifensive escape locomotion in response to blue-light stimulation.

Comments

The expression of paraGD3392 under the control of Scer\GAL4elav.PU, but not of Scer\GAL4Mhc.PU, under 25[o]C temperature, leads to strong decreases in wing posture defects and in the movement time spent in the centre of a circular space exhibited by paraShu heterozygous females.
