No obvious phenotypes are seen when expression of GclcGD9767 is driven by Scer\GAL4GMR.PFa (adult eye) or Scer\GAL4pnr-MD237 (dorsal thorax).

Expression of GclcGD9767 driven by Scer\GAL4elav-C155 leads to developmental arrest at the pupal stage or earlier, with only 19% of flies reaching pupation; rare adult survivors display a sunken thorax with a cleft and unexpanded wings. Larval survival at 96hr post egg hatching is about 56% of that seen in controls when expression of GclcGD9767 is driven by Scer\GAL4elav-C155.

Expression of GclcGD9767 driven by Scer\GAL4CCAP.PP leads to a fully penetrant unexpanded wing phenotype, along with a softened and clefted thoracic cuticle. Scer\GAL4CCAP.PP>GclcGD9767 third instar larval brains show a significant decrease in the density of axon branching compared to controls; though the number of Scer\GAL4CCAP.PP neurons is unchanged, their area is significantly smaller than controls.

Adults expressing GclcGD9767 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Co-expression of ATP7Scer\UAS.T:Zzzz\FLAG or Ctr1AGD16726 significantly enhances larval lethality seen with expression of GclcGD9767 driven by Scer\GAL4elav-C155. Co-expression of ATP7GD3322 or Ctr1BGD2859 does not significantly enhance larval lethality seen with expression of GclcGD9767 driven by Scer\GAL4elav-C155. Co-expression of ATP7Scer\UAS.T:Zzzz\FLAG significantly exacerbates the decrease of axon branching and Scer\GAL4CCAP.PP neuron area in third instar larval brains with expression of GclcGD9767 driven by Scer\GAL4CCAP.PP.
