Description

Imprecise excision of the progenitor P{EPgy2}EndoBEY00696 insertion, resulting in a deletion of sequence encoding the N-terminal 325 amino acid residues of EndoB.

Comment:

Inferred boundaries of a deletion resulting from imprecise excision of P{EPgy2}EndoBEY00696, which removes 324 amino acids of EndoB including the entire BAR domain and the beginning of the SH3 domain. Based on the reported length of EndoB and the relation of the deletion to protein domains, the deletion was surmised to have been reported relative to EndoB-PB.

Detailed Description

Homozygous and EndoB54/Df(2R)BSC26 females lay fewer eggs per day than wild type. The distribution of egg chamber stages in the mutant ovaries is different than wild type,.

Mutant oocytes show a dramatic decrease in the density of yolk granules at their centre compared to wild-type controls.

The high level of endocytosis normally seen at the posterior of the oocyte is not seen in homozygous oocytes.

endoB54 homozygotes exhibit mild lethality.

The fraction of oocytes showing endocytic activity is reduced compared to wild type in EndoB54/Sec5E10, EndoB54/Sec5E13 and EndoB54/Sec6Ex15 double heterozygotes.

The fraction of oocytes showing endocytic activity in EndoB54/Rbsn-5C241 double heterozygotes is not reduced compared to either single heterozygote.

Comments

Expression of EndoBScer\UAS.P\T.T:Zzzz\FLAG under the control of Scer\GAL4unspecified rescues the reduced fecundity of EndoB54/Df(2R)BSC26 females. The altered distribution of yolk granules which is seen in EndoB54 mutant oocytes is also rescued.

Expression of EndoBΔSH3.Scer\UAS.P\T.T:Zzzz\FLAG under the control of Scer\GAL4unspecified rescues the reduced fecundity of EndoB54/Df(2R)BSC26 females.
