Reported amino acid change:

W285

Comment:

G to A nucleotide change at the second or third position of the wild type Trp codon leads to a nonsense mutation (exact site of mutation unspecified). The mutation was annotated at the second base of the codon.

Embryos derived from fzGL31, fz2e2 female germline clones that are also zygotically mutant for fzGL31 and fz2e2 exhibit defects in the specification of naked cuticle. Expression of one copy of the fzαTub84B.PP transgene restores normal patterning to the cuticle of fzGL31; fz2e2 embryos from germline clones and allows 11% of these mutants to eclose. In fz57.αTub84B, fz81.αTub84B and fzΔCRD1.αTub84B rescue crosses, denticles are present in regions that should be entirely naked and some denticle belts are fused; these transgenes are incapable of restoring viability to fzGL31, fz2e2 embryos. fzGL31, fz2e2 clones in the wing often fail to generate bristles altogether. Expression of fzαTub84B.PP in fzGL31, fz2e2 clones in the wing fully restores bristle patterning. fzGL31, fz2e2 clones in the wing are partially rescued by expression of fz57.αTub84B, but still show subtle defects in the spacing of the margin bristles. Clones rescued by fz81.αTub84B show more severe bristle spacing problem. Clones rescued by fzΔCRD1.αTub84B have ectopic margin bristles, but these appear to be derived from neighbouring cells as the bristles are dark, while the clones lack y. Clones rescued by fzWIFCRD.αTub84B still show observable defects in the patterning of the margin, with some places where a bristle socket has formed in the place of an apparently missing bristle.
