Expression of Mmp2Scer\UAS.cPa under the control of Scer\GAL4Mef2.PU does not lead to a significant decrease in the migration velocity of embryonic cardioblasts, nor the number of lamellipodia at the cardioblast leading edge, but the number of filopodia at the leading edge shows a small but significant decrease, the regular cardioblast alignment is disrupted, and multiple ectopic lumens form at the heart proper region, as compared to wild type.

In females with expression of Mmp2Scer\UAS.cPa driven by Scer\GAL4GMR47A04, mature eggs rupture and most lack covering follicle cells.

Scer\GAL4repo.PL + Scer\GAL4repo.PU-mediated expression of Mmp2Scer\UAS.cPa, activated at the onset of third instar larval stage via use of Scer\GAL80ts.αTub84B, causes shape changes in the ventral nerve cord and deformation of the brain lobes 40h later. When induced from first instar onwards, viability is not affected and a more dramatic nervous system phenotype is produced.

Scer\GAL4c527-mediated expression of Mmp2Scer\UAS.cPa results an elongated ventral nerve cord. There is no loss of the ECM, but a rather wrinkled appearance.

Expression of Mmp2Scer\UAS.cPa under the control of Scer\GAL4C587 (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression to the adult stage) results in fused egg chambers which lack stalks (100% penetrance).

Pan-glial expression of Mmp2Scer\UAS.cPa under the control of Scer\GAL4repo.PU strongly increases ISNb and SNa fasciculation.

Flies expressing Mmp2Scer\UAS.cPa in the female fat body under the control of Scer\GAL4yolk are semi-lethal and infertile with a significantly shortened life span, dying a few days post eclosion at 18-20[o]C.

Scer\GAL4elav.PU-mediated expression of Mmp2Scer\UAS.cPa interferes with embryonic motor axon guidance: ISNb and SNa morphology/path-finding is disrupted, and exhibit phenotypes consistent with increased motor axon fasciculation. Expression via Scer\GAL4repo.PU has similar effects but with higher penetrance; the number and position of repo-positive glia are unaffected. Mis-expression of Mmp2Scer\UAS.cPa in mesoderm or in hemocytes does not result in motor axon phenotypes.

Mmp2Scer\UAS.cPa; Scer\GAL4GMR.PF flies have glassy eyes with flattened ommatidia and the occasional missing or double interommatidial bristles.

The follicle rupture defects characteristic for OambMI12417 mutant females are rescued by ectopic expression of Mmp2Scer\UAS.cPa under the control of Scer\GAL4GMR44E10.

Co-expression of TimpScer\UAS.cPa suppresses the fused egg chamber phenotype seen in females expressing Mmp2Scer\UAS.cPa under the control of Scer\GAL4C587 (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression to the adult stage) and restores stalk cell numbers to normal.

Co-expression of Mmp2Scer\UAS.cPa has no effect on the increase in stalk cell numbers seen in females expressing wgScer\UAS.cHa.T:Ivir\HA1 under the control of Scer\GAL4C587 (using the temperature sensitive Scer\GAL80ts.αTub84B allele to limit expression to the adult stage).

A homozygous fracΔ1 mutant background suppresses the excess fasciculation seen in embryos over-expressing Mmp2Scer\UAS.cPa under the control of Scer\GAL4repo.PU. Furthermore, these embryos display diminished motor axon bundling similar to that observed in fracΔ1 homozygotes.

Expression of Mmp2Scer\UAS.cPa in third instar larvae expressing TimpScer\UAS.cPa under the control of Scer\GAL4btl.PS (by inactivating Scer\GAL80ts.alphaTub84B through heat-shock), largely reverts the air sac primordium defects observed when TimpScer\UAS.cPa is expressed alone.

The eye phenotypes due to Mmp2Scer\UAS.cPa; Scer\GAL4GMR.PF are almost completely suppressed by the presence of one copy of TimpScer\UAS.cPa.
