RanGAPEP1173 homozygous third instar larvae display brain lobes with a normal glossy physical appearance, but there is a significant decrease in the number of central brain neuroblasts only in females, which does not show at second instar or early third instar (i.e. 48h and 72h after larval hatching, respectively), as compared to controls. At the pre-pupal stage (i.e. at 120h afer larval hatching) males also start showing a significant decrease in the number of central brain neuroblasts, as compared to controls. Female third instar neuroblasts show a severe decrease in the frequency of cell division (phospho-H3 and EdU stainings), despite of identical levels of apoptosis (Caspase 3 staining) in the brain.

Both male and female RanGAPEP1173/RanGAP327 show a progressive decrease in the number of central brain neuroblasts from early third instar larval stage into prepupal stage (i.e. at 72h-120h after larval hatching, but not at 48h after larval hatching).

Expression via Scer\GAL4fkh.PH results in strongly penetrant gland shape/lumen ('shepherd's crook') and positioning ('hook') phenotypes in the embryonic salivary gland.
