No apparent effect on adult survival and tissue patterning is seen when FakScer\UAS.T:Ivir\HA1 is expressed under the control of either Scer\GAL4GMR.PU or Scer\GAL4ptc-559.1.

Embryos expressing FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4sca-537.4 show defects in the Fas2-positive longitudinal fascicles. Severe gaps are seen in the lateral-most fascicles and aberrant midline crossing of axons is seen in the medial-most fascicles.

Expression of Fak56DScer\UAS.T:Ivir\HA1 in motor neurons under the control of Scer\GAL4D42 in a Fak56DCG1 background does not affect miniature EJP amplitude.

No significant overgrowth of the neuromuscular junction is seen in third instar larvae expressing FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.PU.

Expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4NP4702 results in a thinner optic stalk than normal.

Expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Mef2.PR results in a potent muscle detachment phenotype in embryos, resulting in the rounding up of somatic muscles. Expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E at 25oC results in wing blisters in 28% of adult females (but no wing blistering is seen in adult males).

Expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E results in wing blisters in the posterior region of the wing. Clones in the wing expressing Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Act5C.PI results in wing blisters.

The increased number of intestinal progenitor cells in mitosis characteristic for adult flies expressing RetScer\UAS.T:Zzzz\FLAG,T:Zzzz\His6 driven by Scer\GAL4esg-NP7397 (combined with tub-Gal80[ts] for temporal control) is suppressed by co-expression of FakScer\UAS.T:Ivir\HA1.

Heterozygous FakCG1 enhances the ommatidial mis-patterning seen in eyes expressing RetGMR.PR. Homozygous FakCG1 causes a smaller eye due to a completely disrupted ommatidial pattern. This enhancement is reversed upon co-expression of either FakScer\UAS.T:Ivir\HA1 or FakY430F.Scer\UAS under the control of Scer\GAL4GMR.PU.

Expression of RetGMR.PR partially suppresses the reduction in eye size seen in flies expressing hidGMR.PG. This suppression is partially reversed when FakScer\UAS.T:Ivir\HA1 is expressed under the control of Scer\GAL4GMR.PU.

Expression of FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU partially suppresses the small eye size induced by expression of one copy of RetMEN2A.GMR.

Expression of FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU suppresses the rough eye phenotype seen upon expression of RetGMR.PR. The reduction in retinal apoptosis is also rescued.

Expression of FakScer\UAS.T:Ivir\HA1 suppresses the partial lethality seen when RetMEN2A.Scer\UAS is expressed under the control of Scer\GAL4ptc-559.1. The proportion of surviving flies rises from 46% to 92%. The wing vein and male genitalia defects seen in adult flies are also suppressed.

Expression of FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU does not have a significant effect on the reduced eye size seen upon expression of hidGMR.PG.

Expression of FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PU partially suppresses the eye size defect and ectopic cone cell phenotype induced by expression of two copies of RetMEN2A.GMR, resulting in the reappearance of normal ommatidial cores and surrounding interommatidial cells in pupal retinas.

Expression of FakScer\UAS.T:Ivir\HA1 does not suppress the eye phenotypes seen when phlScer\UAS.F179 is expressed under the control of Scer\GAL4sev.PU.

Expression of FakScer\UAS.T:Ivir\HA1 suppresses the reduction in eye size seen in flies expressing EgfrScer\UAS.cBa under the control of Scer\GAL4GMR.PU.

Expression of FakScer\UAS.T:Ivir\HA1 partially suppresses the almost total lethality seen when EgfrScer\UAS.cBa is expressed under the control of Scer\GAL4ptc-559.1. The proportion of surviving flies is increased to approximately 60%.

The defects seen in the Fas2-positive longitudinal fascicles of embryos expressing FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4sca-537.4 are partially suppressed if the embryos are also heterozygous for Sema-1ak13702.

The penetrance of the wing blistering phenotype seen in females expressing Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E at 25oC is increased from 28% to 44% and 40% if they are carrying mys8/+ or mysb43/+ respectively. Males expressing Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E at 25oC and hemizygous for either mys8 or mysb43 show 31% and 21% wing blistering respectively (compared to 0% wing blistering in a mys+ background).

The inhibition of border cell migration seen in the ovaries of jara.Scer\UAS; Scer\GAL4slbo.2.6 flies is significantly suppressed by the presence of shghs.PW under heat shock condition (flies raised at 29oC and given a 1 hour heat shock at 37oC 6-10 hours prior to dissection.). Partial suppression is also seen in flies carrying slboScer\UAS.cLa or shgScer\UAS.cSa, but not Fak56DScer\UAS.T:Ivir\HA1.

Comments

Expression of Fak56DScer\UAS.T:Ivir\HA1 in motor neurons under the control of Scer\GAL4D42 restores normal glutamate sensitivity to Fak56DCG1 mutants.

Expression of Fak56DScer\UAS.T:Ivir\HA1 in motor neurons under the control of Scer\GAL4D42 restores wild-type levels of long term facilitation in Fak56DCG1 mutants.

The neuromuscular junction defects seen in FakN30/FakK24 larvae are rescued by expression of FakScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.PU, but not under the control of Scer\GAL4Mhc.PW.

The optic stalk defects of Fak56DCG1 homozygotes are rescued by expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4NP4702, partially rescued by expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4NP2109 and are not rescued by expression of Fak56DScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PY.
