The expression of CadNUAS.cIa under the control of Scer\GAL4bsh.M.PT results in the Mi1 neuron terminals in the larval medulla redirecting to the column center and overlapping with R8 photoreceptor terminals, instead of projecting to the grid-like domain outside the columns; the same was observed upon clonal expression of in R7 photoreceptors.

The expression of CadNUAS.cIa under the control of Scer\GAL4sens.F2 results in the R8 photoreceptor terminals in the larval medulla expanding and redirecting to the center of the column and the shrinkage of the central hole area; R7 photoreceptor terminals are also expanded toward the column periphery, overlapping with R8 photoreceptor terminals.

The expression of CadNUAS.cIa under the control of Scer\GAL4sev.PM181 does not lead to significant changes in R7 photoreceptor axon projections in the larval medulla.

Expression of CadNScer\UAS.cIa under the control of Scer\GAL4GH146 does not suppress the dendrite targeting defects seen in homozygous meigo1 projection neuron clones.

Clones in the eye that overexpress CadNScer\UAS.cIa under the control of Scer\GAL4hs.2sev show a rotation defects with ommatidia rotating to a random degree. Random rotation angles are also observed in Scer\GAL4hs.2sev>CadNScer\UAS.cIa third instar eye discs.

Expression of CadNScer\UAS.cIa in CadN405 mutant R7 neurons, under the control of Scer\GAL4tub, increases the percentage of mutant R7 clones with correct target layer selection from 0% to 95%. Expression of CadNScer\UAS.cIa, under the control of Scer\GAL4tub, increases the proportion of wild-type R1-R6 targeting in CadN405 mutant clones.

Clones of CadNScer\UAS.cIa; Scer\GAL4Act5C.PI cells in the pupal retina: When both primary pigment cells in a ommatidium are CadNScer\UAS.cIa; Scer\GAL4Act5C.PI and cone cells are wild-type, the cone cells cluster in a 'cruciform' shape indistinguishable from that due to CadNM19/CadNM19. clones. However, when only one of the two primary pigment cells in an otherwise wild-type ommatidium is CadNScer\UAS.cIa; Scer\GAL4Act5C.PI, there are major changes in cone cell shape and arrangement: the cone cells selectively adhere to the CadNScer\UAS.cIa; Scer\GAL4Act5C.PI primary pigment cell which can be altered into a variety of shapes, often engulfing one or more of the cone cells.

Expression of CadNScer\UAS.cIa under the control of Scer\GAL4GH146 in either neuroblast or single-cell DL1 projection neuron clones (induced using the MARCM technique) do not result in detectable phenotypes in the dendrites of the mutant neurons. There is no change in the axonal branches of the mutant neuron in the mushroom body, but in the lateral horn in 47% of mutant neurons, instead of one dorsal axon branch, there are two dorsal branches.

Expression of CadNScer\UAS.cIa under the control of either Scer\GAL4upd1.PU or Scer\GAL4tj.PU fully rescues the decreased cell numbers of hub cells and cyst stem cells, and partially rescues the decreased number of germline stem cells, in the adult testicular niche of sdaiso7.8/Df(3R)ED6235 mutants.

Expression of CadNScer\UAS.cIa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 fails to rescue the germline stem cell, cyst stem cell, and hub cell number phenotypes in testicular niches of sdaiso7.8/Df(3R)ED6235 mutants.

Expression of CadNScer\UAS.cIa under the control of Scer\GAL4elav-C155 in Lar2127 somatic mutant clones enhances the photoreceptor targeting defects observed in single mutants.

Expression of CadNScer\UAS.cIa under the control of Scer\GAL4elav-C155 in Liprin-α1 somatic mutant clones enhances the photoreceptor targeting defects.

The under-rotation of ommatidia seen in Scer\GAL4hs.2sev>shgdCR3h.Scer\UAS.T:Avic\GFP-rs eyes is enhanced by coexpression of CadNScer\UAS.cIa.

The ommatidial rotation phenotype of Scer\GAL4hs.2sev>CadNScer\UAS.cIa mutants is partially suppressed in an arm4/+ background but gaps appear between ommatidia due to decreased adhesion. Coexpression of armS2.Scer\UAS.T:Hsap\MYC enhances the ommatidial rotational defects of Scer\GAL4hs.2sev>CadNScer\UAS.cIa mutants and causes the loss of some photoreceptors.

Comments

The mistargeting and growth cone morphology defects seen in CadN405 mutant R7 cells at 17% and 35% APF are substantially rescued by expression of CadNScer\UAS.cIa under the control of Scer\GAL4elav-C155.

The abnormal phenotypes seen in the antennal lobe, mushroom body calyx and lateral horn in homozygous CadNM19 projection neurons are rescued by expression of CadNScer\UAS.cIa under the control of Scer\GAL4GH146.
