Detailed Description

stck3R-17/+ does not significantly affect resistance to ethanol-induced sedation.

stck3R-17/stck18 adults rescued by stckD303V.T:Zzzz\FLAG are recovered at 90% of the expected frequency. The rescued adults frequently have wing blisters and wings are often seen in a "held up" position.

Embryos expressing stckwt.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background do not display any muscle structure or actin cytoskeleton defects.

Embryos expressing stckQ38A.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background do not display any muscle structure or actin cytoskeleton defects.

stck3R-17 dorsal branch terminal cell clones do not cause any detectable defects in terminal cell morphology.

Embryos derived from stck3R-17 germ-line clones and zygotically mutant for stck3R-17 display dorsal closure defects, ventral patterning defects and/or head involution defects.

Embryos derived from females carrying stck3R-17 germ-line clones have cuticles with dorsal puckers (36%), dorsal holes (23%), or wild type morphology (12%). Cuticles from embryos both maternally and zygotically stck3R-17 mutant show the same array of phenotypes.

Actin filament arrays are largely retracted to one end of the muscle in stck3R-17/Df(3R)097 late stage 17 embryos. Homozygous clones result in blisters in the wing.

Homozygotes die as early larvae. Homozygous clones in the wing produce discrete, round blisters of variable size. These blisters can be located anywhere on the wing. Wing venation is normal. Homozygous embryos show excessive contractions of the segmental muscles just before hatching. Hatched larvae fail to grow.

Homozygous clones in the wing produce a blistered phenotype.

stck3R-17/+ significantly suppresses (to wild type levels) the increased resistance to ethanol-induced sedation seen in flies with expression of icsdsRNA.Scer\UAS.WIZ driven by Scer\GAL4elav.PU.

flwG0172 ; stck3R-17 /+ animals show complete larval lethality (only a limited number of animals were of this genotype could be isolated due to a high rate of balancer breakdown in the required crosses).

icsBG02577 has no effect on the viability of flies expressing stckwt.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background.

The addition of icsBG02577 reduces the viability of flies expressing stckQ38A.T:Zzzz\FLAG in a stck3R-17/stck18 mutant background. Between 6 and 34% of the expected number of progeny are seen, with the majority of the lethality taking place during the larval and pharate stages. The rescued larvae display a range of sizes, including presumptive first instar larvae that appear stumpy, exhibit sluggish movement and do not progress in development.

Shows a weak genetic interaction with mys8 and mysb9; the frequency of held-out wings but not the frequency of blisters in mys8 or mysb9 single hemizygotes is increased if they are also heterozygous for stck3R-17.

Comments

Expression of stckwt.T:Zzzz\FLAG rescues the embryonic and larval lethality seen in stck3R-17/stck18 mutants. The rescued flies do not display any overt phenotypes.

Expression of stckQ38A.T:Zzzz\FLAG rescues the embryonic and larval lethality seen in stck3R-17/stck18 mutants. The rescued flies are fertile and do not display either wing blisters or any other overt phenotypes.
