awdKRS6 females in which the lethality has been rescued by expression of awdhs.P at 29[o]C and then the females have been shifted to 18[o]C for 3 days have very few intact egg chambers. The few surviving egg chambers are reduced and uneven and show disrupted epithelial organisation and epithelial discontinuity.

awdMSM95/awdKRS6 flies are paralysed at 38oC. These flies lose the on- and off-transients normally seen in an electroretinogram at 39oC in a use dependent manner. At 39oC, membrane invaginations are seen at a low level in the photoreceptor terminals in the lamina in awdMSM95/awdKRS6 flies.

Ovaries from homozygous larvae transplanted into female sterile host larvae fail to develop egg chambers. 21 of 26 egg laying females generated from embryos that had been transplanted with awdKRS6 pole cells gave rise to viable progeny, the females are heterozygous. The remaining 5 females are homozygous and give rise to embryos that survive as late as the blastoderm stage. Surprisingly females carrying germline clones give rise to viable progeny, the only source of awd present is in the cytoplasm of the heterozygous clone mother cells.

The temperature at which the ERG on- and off-transients are lost is lowered to 30oC in shiCK2 flies that also carry awdMSM95/awdKRS6. The temperature at which the membrane invaginations are seen in the photoreceptor terminals in the lamina in shiCK2 flies is reduced if they are also carrying awdMSM95/awdKRS6. shiCK2/Y ; awdMSM95/awdKRS6 motor terminals show reduced uptake of an endocytosis tracer at 38oC compared to controls. The general morphology of the motor nerve endings appears normal in these flies.

Hsap\NME1awd.T:Ivir\HA1 exhibits partial rescue of the awdKRS6 mutant zygotic phenotype, a greater degree of rescue is seen with Hsap\NME2awd.T:Ivir\HA1.
