Description

Deletion of 33 nucleotides associated with an insertion of 13 nucleotides, 370 amino acids downstream from the start of translation. This causes a frameshift mutation and premature stop codon within the coding region.

m1 hemizygotes show a small wing phenotype.

Mutant larvae have thick, broad denticles.

Each denticle row is present, but the denticles themselves are small and lack the median constriction, adopting an abnormal triangular shape.

Mutants show a strong reduction in the size of adult wings.

m1 embryos have impaired contacts between the denticle membrane and cuticle layers

m1 larvae have small, misshapen denticles that display an aberrant triangular shape. Additionally, the denticle cells do not contact cuticle correctly.

m68/m1 females have a m mutant phenotype and are fertile.

m1/In(1)mK females may or may not show the m mutant phenotype, some being normal or almost wild-type.

Wing size reduced, only slightly longer than abdomen and with normal proportions. Angle between L2 and L5 reduced. Wings dark gray and less transparent than normal. Wing cells smaller than normal (Dobzhansky, 1929). In poor cultures, wings may become divergent and stringy. Cell expansion inhibited in prepupae and pupae [Waddington, 1940). RK1.

m1 ; ckKT9 double mutants show a reduction in both size and sharpness of the denticles compared to either single mutant.

m1 f1 ckKT9 triple mutants show almost no elongation of the denticles.

m1, tynPG38 double mutants have denticles with thin extremities (as found in tynPG38 single mutants) and lacking the medial constriction (as found in m1 single mutants). The addition of Df(3L)zyeex72, which deletes zye, has a further effect, leading to denticles with a twisted appearence.

Expression of dylScer\UAS.cFa with Scer\GAL4da.G32 slightly modifies the m1 denticle phenotype, but doesn't rescue m1 the adult wing phenotype.

Expression of m::tynPAN-tyn.Scer\UAS with Scer\GAL4da.G32 rescues the m1 denticle phenotype and the m1 adult wing phenotype.

Expression of m::tynZPtyn.Scer\UAS with Scer\GAL4da.G32 fails to rescue the m1 denticle phenotype and only partially restores the m1 adult wing phenotype.

Expression of m::tynZPm.Scer\UAS with Scer\GAL4da.G32 has a weak effect on the m1 denticle phenotype and significantly rescues the m1 adult wing phenotype.

The denticles of m1, sn3 mutants show splitting in addition to the defects seen in either single mutant. m1, f36a mutants also show denticle splitting. sn3, m1, f36a triple mutants have small, highly misshapen denticles that are more affected than those of m1, f36a or m1, sn3 double mutants. sn3, m1, f36a; shaV15 and sn3, m1, f36a; WASp3 quadruple mutants show further impairment to denticle formation compared to sn3, m1, f36a triple mutants, with mutants exhibiting regions of naked cuticle where denticles lie in wild-type animals.

The dorsal hairs on the abdominal segments of sn3, m1, f36a triple mutants are severly reduced in size, and in some cases, hairs are abrogated leaving abnormal naked regions. This phenotype is more severe in sn3, m1, f36a; WASp3 quadruple mutants and is even more severe in sn3, m1, f36a; shaV15 mutants in which most dorsal hairs are absent, leaving naked cuticle.

Expression of mScer\UAS.cFa or mScer\UAS.T:Arus\HRP using Scer\GAL4da.G32 rescues the denticle and adult wing size phenotypes of m1 animals.

Expression of mScer\UAS.cFa using Scer\GAL4da.G32 rescues the denticle defects of m1 embryos and the reduction in wing size of m1 adults.
