Imprecise excision of P{SUPor-P}Rgk1KG00183 results in deletion of part of Rgk1. The first exons of the RA and RC isoforms are absent and there is partial deletion of the RB isoform exon. There is a frameshift in the RB isoform at 3405 bp leading to a change of the translated peptide (QYV to HPR) followed by a stop codon, resulting in a truncated Rgk1 without the putative GTPase domain.
Rgk1Δ homozygous adults and Rgk1Δ/Df(2R)BSC26 transheterozygous adults, but not either Rgk1Δ or Df(2R)BSC26 heterozygotes, display a significant decrease in intermediate-term olfactory memory (2h after conditioning), assessed by olfactory aversive conditioning assays, which is observed regardless of the individuals being subjected to cold-shock anesthesia or not, as compared to controls; Rgk1Δ homozygous adults also display a small but significant decrease in short-term olfactory memory (5 min), as compared to controls. None of the above allelic combinations leads to defects in olfactory acuity, response to electric shocks, or morphology of the mushroom body, as compared to controls.

Adult males that are both homozygous for Rgk1Δ and hemizygous for either dnc1 or rut2080 exhibit a severe decrease in cold-shock anesthesia-resistant intermediate-term olfactory memory (3h after conditioning) as compared to adult males that are only homozygous for Rgk1Δ or, respectively, only hemizygous for dnc1 or rut2080.
Heterozygosity for Rgk1Δ enhances the intermediate-term olfactory memory (3h after conditioning) defects resulting from the adulthood-only expression of Rac1V12.Scer\UAS under the combined control of Scer\GAL4ey-OK107 and Gal80[ts].

The expression of Rgk1Scer\UAS.B, under the control of Scer\GAL4ey-OK107 alone, or in combination with Gal80[ts] to transiently express only for 3 days during adulthood, rescues the intermediate-term olfactory memory (2h after conditioning) defects observed in Rgk1Δ homozygous adults, and even leads to significant increases in intermediate-term memory performance as compared to controls; expression under the control of Scer\GAL4c739 alone or Scer\GAL4elav-C155 alone also partially and fully rescues, respectively, the intermediate-term olfactory memory defects of Rgk1Δ homozygous adults.
The expression of either Rgk1Scer\UAS.B.T:Avic\GFP or Rgk1ΔN.Scer\UAS.B.T:Avic\GFP, but not of either Rgk1ΔC.Scer\UAS.B.T:Avic\GFP or Rgk1S1134N.Scer\UAS.B.T:Avic\GFP, under the control of Scer\GAL4ey-OK107 rescues the intermediate-term olfactory memory defects observed in Rgk1Δ homozygous adults, and even leads to a small increase in intermediate-term memory performance, as compared to controls; the rescue by Rgk1Scer\UAS.B.T:Avic\GFP, but not by Rgk1ΔN.Scer\UAS.B.T:Avic\GFP, even occurs if the individuals are subjected to cold-shock anesthesia after conditioning.
