Description

Deletion of approximately 31kb of genomic sequence that removes the predicted translation start site of all Eip78C isoforms and the DNA-binding domain of isoforms RA/RD/RE. Generated by FLP-mediated recombination between the two progenitor insertions (PBac{RB}e02923 and PBac{RB}Eip78Ce02853).

Eip78CΔ31/Eip78CΔ31 animals are viable and male fertile but completely female sterile.

The number of eggs laid by the Eip78CΔ31/Df(3L)BSC419 transheterozygote females as well as the proportion of laid eggs that actually hatch is significantly decreased compared to age-matched controls. Majority of embryos derived from Eip78CΔ31 females fails to gastrulate.

Eip78CΔ31/Df(3L)BSC419 transheterozygotes show increased developmental lethality when raised on a sugar-rich (K12 media) diet, which can be rescued by addition of yeast paste.

Eip78CΔ31/Df(3L)BSC419 transheterozygote females have on average fewer germline stem cells (GSC) per germarium and show higher proportion of germaria containing less than three GSCs compared to age-matched controls. However, the rate of proliferation of GSCs and their maintenance over time is not perturbed in Eip78CΔ31 mutants compared to controls.

Eip78CΔ31/Df(3L)BSC419 transheterozygote females show slight but significant decrease in the average number of cap cells per germarium, regardless of age of the female.

The proportion of ovarioles containing dying follicles (both early and late stages) is highly increased in Eip78CΔ31/Df(3L)BSC419 transheterozygote females compared to wild-type as well as in mosaic animals with ovarioles containing Eip78CΔ31/Eip78CΔ31 germline clones. Stage 4/5 Eip78CΔ31 mutant cysts are significantly overrepresented, while stage 8-10 mutant cysts are significantly underrepresented in ovarioles containing Eip78CΔ31 germline clones compared to controls.

The levels of follicle death (scored as percentage of ovarioles containing at least one dying follicle) in Eip78CΔ31;EcRM554fs double heterozygote females is significantly increased compared to either Eip78CΔ31/+ or EcRM554fs/+ alone.

The levels of follicle death (scored as percentage of ovarioles containing at least one dying follicle) in Eip78CΔ31;ecd1 double heterozygote females is significantly increased compared to either Eip78CΔ31/+ or ecd1/+ alone.

The levels of follicle death (scored as percentage of ovarioles containing at least one dying follicle) in Eip78CΔ31;Eip75BΔ51 double heterozygote females is not significantly increased compared to either Eip78CΔ31/+ or Eip75BΔ51/+ alone.

Comments

The decreased number of cap cells per germarium and the high proportion of ovarioles that contain dying follicles characteristic for Eip78CΔ31/Df(3L)BSC419 adult females can be partially restored by combination with single copy of Eip78CT:Avic\GFP-SF,T:Zzzz\FLAG but the reduced number of germline stem cells per germarium is not rescued.
