Description

Deletion of residues 220-460 of the ktub coding region (this includes sequence encoding the conserved "tubby" domain). A w+mW.hs marker has been inserted in place of the deleted ktub sequence.

ktub1/ktub1 or ktub1/ktubG flies show significant locomotor defects (reduced climbing ability) compared to wild type. ktub1/ktub1 or ktub1/Df(2R)BSC462 flies show a significant reduction in sound evoked potentials from the antennal nerve compared to wild type. ktub1/ktub1 flies show an overall normal Johnston's organ structure, though electron microscopy reveals an abnormal number of chordotonal cilia (in around 6% of scolopidia) and cilia are found outside the cap structure (in around 3% of scolopidia).

INPP5EMI02221/INPP5EMI02221 ktub1/ktub1 flies with expression of ktubScer\UAS.cPa or ktubmutB.Scer\UAS driven by Scer\GAL4nan.PK show significantly reduced amplitude of sound-evoked potentials from antennal nerves compared to ktub1/ktub1 Scer\GAL4nan.PK>ktubScer\UAS.cPa flies (similar to wild type amplitude); however, INPP5EMI02221/INPP5EMI02221 ktub1/ktub1 flies with expression of ktubmutB.Scer\UAS have a significantly increased amplitude in comparison to those with expression of ktubScer\UAS.cPa.

Comments

Expression of ktubScer\UAS.cPa (but not ktubmutB.Scer\UAS) driven by Scer\GAL4nan.PK leads to amplitude of sound-evoked potentials from ktub1/ktub1 antennal nerves being similar to controls.

ktub+tCH321-59C17 rescues locomotor and auditory defects in ktub1/ktub1 flies. Expression of ktubScer\UAS.cPa fully rescues, ktubmutA.Scer\UAS or ktubmutB.Scer\UAS partially rescues, and ktubmutAB.Scer\UAS fails to rescue auditory defects in ktub1/ktub1 flies when driven by Scer\GAL4nan.PK .
