Eyes composed entirely of lin-5293 mutant clone cells (generated using the ey-FLP method) are rough and include small rhabdomeres and irregular ommatidia. When lin-5293 mutant eye clones are generated alongside wild type control tissue mutant the mutant ommatidia are disorganised and contain rhabdomeres that are distended and/or absent compared to the adjacent control cells. No defects are observed in the control cells, indicating that the phenotype is cell-autonomous.

Less than 1% of embryos derived from females carrying homozygous lin-5293 germline clones crossed to wild type males are able to hatch. in 86% of embryos mitotic arrest occurs when the embryo contains between two and eight nuclei. In many of these embryos the mitotic spindles and metaphase plate have failed to form correctly, with condensed chromosomes often distributed diffusely between the spindle poles. One or both centrosomes are often missing and other defects observed include a broad spindle with no centrosomes, a branched spindle with no centrosomes or an elongated spindle with no centrosomes.

Generation of lin-5293 mutant clones in the germline or ovarian somatic cells has no effect on the morphology of these cells, nor on the progression of oogenesis.

mip40EY16520 rescues the lethality seen in homozygous and hemizygous lin-5293 larvae. Both male and female adult flies are seen at a similar frequency to controls. One copy of mip40EY16520 is not sufficient to rescue the lethality.

Homozygous mip1301-36 suppresses the eye phenotypes seen in lin-5293 mutant eye clones. The mutant tissue appears completely normal.

Homozygous MybMH107 does not suppress the eye phenotypes seen in lin-5293 mutant eye clones.

Homozygous mip1301-36 suppresses the eye phenotypes seen in MybMH107, lin-5293 double mutant clones. The resulting triple mutant eyes appear completely normal.

Approximately 29% of embryos derived from females carrying homozygous lin-5293 mip1301-36 double mutant germline clones crossed to wild type males are able to hatch, a partial suppression of the phenotype seen when females are maternally mutant for lin-5293 alone.

Approximately 2% of embryos derived from females carrying homozygous lin-5293 MybMH107 double mutant germline clones crossed to wild type males are able to hatch, a similar rate to when females are maternally mutant for lin-5293 alone.

Embryos derived from females that carry homozygous lin-5293 mutant germline clones and are heterozygous for mip40EY16520 crossed to wild type males are unable to hatch, similar to when females are maternally mutant for lin-5293 alone.

Approximately 21% of embryos derived from females that carry homozygous lin-5293 mutant germline clones and are heterozygous for mip12067 (Df(2R)mip12067 mutants in which EfTuM[+] function has been restored using EfTuM+tBa) crossed to wild type males are able to hatch, a partial suppression of the phenotype seen when females are maternally mutant for lin-5293 alone.
