A limited number of adult flies can be obtained when Cul-3NIG.11861R is expressed under the control of Scer\GAL4Clk.8424.
Under light-dark conditions, flies expressing Cul-3NIG.11861R under the control of either Scer\GAL4P2.4.Pdf or Scer\GAL4Clk.8424 show no or reduced morning anticipatory activity. After transfer to constant darkness conditions, the flies either show weak locomotor activity rhythms or become arrhythmic.
Flies expressing Cul-3NIG.11861R under the control of Scer\GAL4P2.4.Pdf which are raised at 25[o]C and tested at 20[o]C show weaker locomotor activity rhythm defects under constant darkness conditions than flies raised and tested at 25[o]C, indicating that Cul-3 function is required during the adult stage for normal locomotor activity rhythms.
Flies expressing Cul-3NIG.11861R under the control of Scer\GAL41118 show reduced morning anticipation under light-dark conditions. Under constant darkness conditions, only 44% of the flies are rhythmic for locomotor activity and the rhythmic flies display weak rhythms.
Under constant darkness conditions, only 30% of flies expressing Cul-3NIG.11861R under the control of Scer\GAL4cry.PE are rhythmic for locomotor activity and the rhythmic flies display weak rhythms.
Expression of Cul-3NIG.11861R under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi effects) results in sleep defects: the flies show a reduction in sleep duration compared to controls, with a reduction in the average sleep bout length and an increase in latency to sleep after lights-off. The mutant flies show defects in sleep homeostasis, lacking a detectable sleep rebound after sleep deprivation. The mutant flies are more easily woken by a rotational stimulus compared to wild type controls.
Expression of Cul-3NIG.11861R under the control of Scer\GAL430Y (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi effects) has no affect on sleep.
Expression of Cul-3NIG.11861R under the control of Scer\GAL4elav.Switch.PO during development (using RU486 to control the timing of expression and in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi effects) results in reduced sleep duration, while expression of incKK108908 under the control of Scer\GAL4elav.Switch.PO during the adult stage only does not result in sleep defects.
Expression of Cul-3NIG.11861R under the control of Scer\GAL4elav-C155 results in a small decrease in sleep duration. In the presence of Dcr-2Scer\UAS.cDa (to enhance the strength of RNAi) these animals show a severe decrease in sleep duration and bout length compared to controls.
Co-expression of both incKK108908 and Cul-3NIG.11861R under the control of Scer\GAL430Y (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi effects) results in a significant decrease in sleep duration and average sleep bout length.
Cul3NIG.11861R is partially rescued by Scer\GAL4elav.PLu/Cul3UAS.cMa
Co-expression of Cul-3Scer\UAS.cMa partially rescues the mutant sleep phenotypes caused by expression of Cul-3NIG.11861R under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to enhance RNAi effects).
