Overexpression of atlScer\UAS.T:Hsap\MYC driven by Scer\GAL4arm.PS does not affect mEJP amplitude at the NMJ of third instar larvae.

Overexpression of atlScer\UAS.T:Hsap\MYC driven by Scer\GAL4Toll-6-D42 causes accumulation of large endoplasmic reticulum punctae in motor neuron cell bodies and axons in third instar larvae; excitatory junction potential (EJP) amplitude is significantly decreased (at 0.1 mM Ca[2+], accompanied by a significant decrease in frequency of synaptic transmission successes); corrected quantal content at 1.5 mM Ca[2+] significantly decreases compared to controls at the neuromuscular junction of third instar larvae.

Expression of atlScer\UAS.T:Hsap\MYC under the control of Scer\GAL4ey.PH or Scer\GAL4GMR.PF results in a rough eye or a small eye phenotype, respectively.

Ultrastructural analysis shows considerable disruption of endoplasmic reticulum (ER) networks in neurons as a result of atlScer\UAS.T:Hsap\MYC] overexpression driven by Scer\GAL4D42. Normal tubular ER profiles are absent in atlScer\UAS.T:Hsap\MYC] overexpressing motoneurons where ER membranes form expanded cisternae.

The small eye phenotype characteristic for adult flies expressing atlScer\UAS.T:Hsap\MYC under the control of Scer\GAL4GMR.PF is exacerbated further by co-expression of TER94Scer\UAS.cRa and is significantly ameliorated by combination with a single copy of any of the following alleles: TER9403775, TER9422-30, TER94k15502 or TER9426-8. The rough eye phenotype of flies expressing atlScer\UAS.T:Hsap\MYC driven by Scer\GAL4ey.PH is also worsened by TER94Scer\UAS.cRa co-expression leading to rough eyes of reduced size.
