Although mean cell number increases in MARCM clones expressing Scer\GAL4tub.PU>tkvca.Scer\UAS.P\T 24 hours after bleomycin treatment, the tkvca.Scer\UAS.P\T-expressing clones have fewer cells than wild-type clones after 48 and 72 hours.

Expression of tkvca.Scer\UAS.P\T in germline cells using Scer\GAL4nos.UTR.T:Hsim\VP16 results in the germaria containing an average of 2.6 cystoblasts per germanium, in comparison with about one cystoblast per wild-type germarium.

The germaria of flies expressing tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 contain an increased number of spectrosome-containing GSC-like cells.

Expression of tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 in females results in a tumorous germarium consisting of undifferentiated germline stem cell-like cells that carry spectrosomes.

Lis-1D heterozygous germaria overexpressing tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 exhibit significantly fewer cystoblasts than wild-type germaria overexpressing tkvca.Scer\UAS.P\T.

Expression of fuScer\UAS.P\T.T:Myr-Src64B partially suppresses the increase in spectrosome-containing GSC-like cells seen when tkvca.Scer\UAS.P\T is expressed under the control of Scer\GAL4nos.UTR.T:Hsim\VP16. The germaria contain branched fusomes and 30% of ovarioles carry normal egg chambers.

Co-expression of lackScer\UAS.P\T.cXa and fuScer\UAS.P\T.T:Myr-Src64B strongly suppresses the increase in spectrosome-containing GSC-like cells seen when tkvca.Scer\UAS.P\T is expressed under the control of Scer\GAL4nos.UTR.T:Hsim\VP16. The germaria contain branched fusomes and nearly 50% of egg chambers contain normal egg chambers.

Expression of tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 suppresses the loss of germline stem cells which is seen in the ovarioles of dallygem females.

Introduction of one copy of Oteunspecified into females expressing tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 results in some of the germline stem cell-like cells in the ovaries differentiating into cyst cells with highly branched fusomes. The germaria are attached with a long string of egg chambers and fertility is restored.

Females expressing tkvca.Scer\UAS.P\T under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 in a homozygous Oteunspecified background have a phenotype that is similar to that found in Oteunspecified single mutants; the majority of germaria are either empty or contain only differentiated germ cells.
