Expression of PuraGD12260 under the control of Scer\GAL4P0.5.Pdf (limited to the adult stages using Scer\GAL80ts.αTub84B) results in an increase in axonal volume and spread of s-LNv neuron projections compared to controls when expression is induced at ZT24 (dawn) and spread is measured 12 hours later. The spread of projections is unaffected when expressed is induced at ZT12 (dusk) and measured at dawn. Normal locomotor activity is seen when the flies are kept in complete darkness (DD conditions).

Expressing PuraGD12260 for 14 hours under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B) has no effect on the number of active zones in LNv projections at ZT2, but the normally low number of active zones seen at ZT14 is increased to the levels seen at ZT2 in controls.

Adults expressing PuraGD12260 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4Mef2.PR results in adult/pharate lethality.

Expression of PuraGD12260 does not suppress the increase in length of the behavioral rhythm period seen when Rac1Scer\UAS.cLa is expressed under the control of Scer\GAL4P0.5.Pdf.

Co-expression of PuraGD12260 suppresses the reduction in axonal volume and spread of s-LNv neuron projections seen at ZT24 (dawn), following 12 hours of expression of Rho1Scer\UAS.cMa under the control of Scer\GAL4P0.5.Pdf (restricted to the adult stages using Scer\GAL80ts.αTub84B).

Expression of PuraGD12260 does not suppress the arrhythmicity seen in flies expressing Rho1V14.Scer\UAS under the control of Scer\GAL4P0.5.Pdf.
