Reported amino acid change:

W189term

Comment:

G to A nucleotide change at the second or third position of the Trp codon leads to a nonsense mutation (exact site of mutation unspecified). Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change.

Detailed Description

Ext21.8.1/Df(2R)Jp6 embryos from Ext21.8.1 germ line clone females fail to hatch. The resulting mature embryonic cuticles indicate a strong segment polarity phenotype: naked cuticle is largely absent, and instead the ventral surface of the cuticle is covered by a near contiguous lawn of denticles. Wings carrying clones of Ext21.8.1 homozygous cells have veination defects effecting veins L3 and L4, notching of the anterior wing margin and ectopic bristles in the vicinity of the wing margin. These ectopic bristles sometimes occur in adjacent heterozygous or wild-type cells. Ext21.8.1/Df(2R)Jp6 third instar larvae contain minute levels of heparin sulphate compared to wild-type. However, levels of chondroitin sulphate are only modestly reduced.

ttv00681b Ext21.8.1 double heterozygotes exhibit increased median survival time compared to controls when infected with group B Streptococcus (both wild-type and GAG-binding impaired mutants) and lower colony counts after infection.

ttv00681b Ext21.8.1 double heterozygotes exhibit reduced susceptibility to infection with the A909 strain of group B streptococcus.

The wing phenotypes due to clones of Ext21.8.1; ttv02055 double homozygous cells are no more severe than those due to single mutant clones of either flavour.
