Clones of Notum3 in pupal eyes show an increase in the number of peripheral ommatidia that undergo apoptosis and a corresponding expansion of the pigment rim.

Homozygous embryos derived from females containing homozygous germline clones have a range of severe defects. Most of these embryos have abnormal morphology; 43% have a posterior end that is not completely retracted, 12% are U-shaped and 34% are shaped like a ball. Of those embryos where the ventral cuticle can be reliably analysed, nearly all have thinner denticle belts in the abdomen than normal, and many have either patches of naked cuticle within the denticle belts, or missing denticle belts. Paternally rescued embryos derived from females containing homozygous germline clones hatch and do not show any morphological defects.

Notum3/NotumI6 larval wing discs show defects ranging from duplication of the wing pouch and hinge associated with a reduced thorax region to almost complete loss of the thorax region associated with a severely abnormal wing duplication. Large clones in the wing disc produce additional rows of sensory organ precursor cells, resulting in extra mechanosensory bristles in the anterior wing margin. These defects only occur when the clone affects both sides of the dorsoventral boundary. Some of the ectopic bristles are derived from wild-type cells neighbouring the clone.

dlp2 +/+ Notum3 transheterozygotes show increased bristle density at the anterior wing margin and ectopic bristles positioned further from the wing margin than in wild type. Neither dlp2/+ or Notum3/+ single mutant flies show this phenotype.
