Expression of grnScer\UAS.cBa in dMP2 neurons, under the control of Scer\GAL4Vap.P0201 has no effect on dMP2 pathfinding.

In most larvae expressing grnScer\UAS.cBa under the control of Scer\GAL4h-1J3, the denticle belts of the alternating segments are reduced or missing, leaving a naked cuticle area. The head skeleton is abnormal in 9-32% of embryos and tail invagination is seen in 6-20%, depending on the number of copies of grnScer\UAS.cBa and the temperature of culture. In embryos with invaginated tails, the spiracles are now at the end of a long tubular structure formed by segments posterior to A8 that normally would be on the outside of the embryo. This structure includes cells from A8, A9, A10 and the telson.

Expression of grnScer\UAS.cBa in the aCC/RP2 neurons, under the control of Scer\GAL4eve.RN2 does not rescue the failure of aCC to project its axon out of the ventral nerve cord in eveΔRP2A mutants.

Expression of grnScer\UAS.cBa and eveScer\UAS.cBa in dMP2 neurons, under the control of Scer\GAL4Vap.P0201 enhances the eveScer\UAS.cBa dMP2 pathfinding defect, resulting in a high frequency of lateral exit out from the ventral nerve cord (in approximately 45% of cases).

Simultaneous expression of upd1Scer\UAS.cZa, emsScer\UAS.cSa, ctScer\UAS.cLa and grnScer\UAS.cBa, under the control of Scer\GAL469B allows formation of spiracle-like tubular structures in Abd-BM1/Abd-BM1 embryos.

Comments

Expression of grnScer\UAS.cBa in the aCC/RP2 neurons, under the control of Scer\GAL4eve.RN2 efficiently rescues grn7L/Df(3R)dsx3 and grnSPJ9/Df(3R)dsx3 mutant axon pathfinding (with 100% muscle 1/9 innervation). By contrast, expression of grnScer\UAS.cBa in the U motoneurons, under the control of Scer\GAL4eve.CQ2 only partially rescues the grn7L/Df(3R)dsx3 and grnSPJ9/Df(3R)dsx3 mutant phenotypes, with approximately 54% of muscles 1/9 being innervated.

Partially rescues the posterior spiracle phenotype of grn7J embryos when expressed under the control of Scer\GAL4salm-459.2.
