Description

The mutated residue R30 is perfectly conserved in homologs from S. cerevisiae to H. sapiens, in a region of the protein that is almost perfectly conserved, and that is thought to be required for deacetylase activity.

Amino acid replacement: R30C.

Detailed Description

Rpd3313/+ flies have the normal number of dorsocentral bristles.

One copy of Rpd3313 strongly suppresses position effect variegation (PEV) at the w locus caused by In(1)wm4.

One copy of Rpd3313 weakly suppresses the telomeric position effect (TPE) in stocks carrying a variegating P{hsp26-pt-T}39C-5 insertion at the telomere of the left arm of chromosome two.

Rpd3313/+ enhances the silencing of the w marker gene which is seen in the P{UAS-lacZ.Abd-B.24F6}GCD-6 insertion, such that the eye is less pigmented in the presence of Rpd3313/+.

Strong suppressor of PEV in In(1)wm4. Raises the pigment levels of In(1)wm4 eyes from 5-15% of wild type levels to 60-90% of wild type levels. Lethality principally affects the third larval instar stage. Shows a dominant semi-lethal effect on males: viability 51% of expected.

Flies heterozygous for Rpd3313 and pnrD1 do not show enhancement of the pnrD1/+ phenotype.

No Rpd3313 mutant males expressing GugdsRNA.1.Scer\UAS under the control of Scer\GAL4dpp.blk1 develop melanotic masses in their heads.

60% of Rpd3313/Df(3R)sbd45 transheterozygous males show transformation of A6 to A5, as judged by the presence of at least one bristle on the sixth sternite.

Comments

Transheterozygotes of Rpd315-1 with Rpd3303, Rpd3313, Rpd3326 or Rpd3328 are viable and fertile, and show weak to moderate suppression of In(1)wm4 PEV.
