Description

An Hsp70 promoter and two copies of the 670bp sev enhancer drive expression of a mutant form of Src42A that carries a K276R amino acid replacement in the ATP-binding site. This results in a dominant-negative form of the protein.

      Eyes of Src42AKR.hs.2sev/+ flies are almost entirely normal.

      Heterozygotes exhibit rough eye phenotype. Mutant phenotype is suppressed by an extra copy of Src42A provided by P{sE2-Dsrc41+} and enhanced by loss of one copy of Src42A, In(2R)bwVDe2LCyR.

      The Src42AKR.hs.2sev/+ eye phenotype is enhanced when flies are also heterozygous for shgE7A-1 or shgR64a. Expression of the Src42AKR.hs.2sev transgene in a arm8 background results in a rough eye phenotype.
