Mlc2E38 mutant flies expressing Mlc2S66A,S67A display decreased wing beat frequency and few of them are able to fly or beat their wings resulting in a large reduction in flight index.

Mlc2E38 mutants carrying Mlc2S66A,S67A display sarcomeric structural abnormalities as revealed by X-ray diffraction and electron microscopy.

Most flies carrying Mlc2S66A,S67A are unable to beat their wings.

The myofibrillar and sarcomeric structures of the indirect flight muscle (IFM) of Mlc2E38 flies carrying Mlc2S66A,S67A appear normal. IFM fibers fixed in the rigor state have well-ordered cross-bridge arrays. The flight index of these flies is significantly less than for Mlc2E38 flies carrying Mlc2+tTa (0.9 +/- 0.6). The wingbeat frequency of these flies is 152 s-1.

Indirect flight muscles appear normal.

Indirect flight muscles appear normal. Two copies of P{Mlc2S66A,S67A} restores viability to Mlc2E38 homozygotes.
