The 2.5kb, 3.7kb, and 5.0kb tws transcripts are nearly absent in brain and imaginal disc RNA from twsaar-1 mutants. Higher molecular weight transcripts are observed which are thought to result from incorrect splicing.

Homozygous or twsj11C8/twsaar-1 flies die as pharate adults or young adults which have duplicated bristles. twsaar-1/Df(3R)by62 flies die as early pupae with duplicated wing imaginal discs at the third larval instar stage, or die as pharate adults with roughened eyes and duplicated bristles.

Lagging chromatids and chromatin bridging are often observed during anaphase in larval brain cells.

Larval neuroblasts have a high mitotic index and chromosome abnormalities at division.

Orcein stained brain preparations reveal mitotic abnormalities: cells, frequently hyperploid have overcondensed chromosomes, anaphases have lagging, bridged or stretched chromosomes. Whole mount preparations revealed abnormal mitotic spindles. In situ hybridizations with centromere and telomere probes showed that DNA replication was completed and sister chromatid separation occurred in lagging chromosomes, but the chromatids had not moved to the spindle poles.

twsaar-1/+ enhances the reduced fertility phenotype of polo1/+ gwlscant/+ double heterozygous females, such that the triple heterozygotes are completely sterile.

The embryos resulting from the cross between polo11/+, twsaar-1 mothers and wild-type fathers fail to hatch.

One copy of twsaar-1 partially suppresses the reduction in eye size seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF. Eyes are 83% of wild type size in females and 74% in males.

One copy each of twsaar-1 and tws60 completely suppresses the reduction in eye size seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4GMR.PF.

One copy of twsaar-1 partially suppresses the wing phenotype seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4en.PU at 29[o]C. Loss of the posterior part of the wing is seen in 8% of flies, compared to 100% when Zzzz\E4orf4Scer\UAS.cPa is expressed alone. Many wings exhibit more minor phenotypes and in 25% of flies the wings appear wild type.

One copy of twsaar-1 partially suppresses the wing disc phenotype seen when Zzzz\E4orf4Scer\UAS.cPa is expressed under the control of Scer\GAL4en.PU at 29[o]C. Only 30% of wing discs contain a cluster of dead cells, compared to 100% when Zzzz\E4orf4Scer\UAS.cPa is expressed alone.
