Lesion in an enhancer element.
A small deletion in the 5' regulatory region of wg.
Regulatory mutation.
1kb deletion located 9kb 5' of the promoter, in the 5' regulatory region, removing an enhancer element.
In wgspd-fg mutant third instar larval wing imaginal discs, the prospective distal hinge region is reduced in size, however, there is no increase in apoptotic cells compared to wild-type.
The wing hinge region is deleted in mutant flies and the wing pouch appears directly joined to more proximal cells.
Homozygotes have shortened wings, alula etching (loss of the margin) and occasional etching of the posterior wing margin. Wings are held upward, the 'tent' phenotype. Transheterozygotes with wg alleles show an allele-dependent extent of alular etching and may show the tent phenotype. Transheterozygotes with null wg alleles only show etching of the alar lobe. Loss of Sternopleural function (by wgSp-1 or Df(2L)RF) exaggerates the etched wing phenotype of homozygotes.
Overall reduction in wing size, reduction of the alula and variable loss of posterior wing margin structures. wgl-17/wgspd-fg flies show a milder version of the hinge phenotype seen in wgspd-fg homozygotes. A deficiency that uncovers wg does not exhibit a hinge phenotype. Heterozygotes with wgspd-j2 produce a hinge phenotype indistinguishable from wgspd-fg homozygotes.
Homozygotes have small wings with severely reduced alula and defects at the wing margin. When placed in trans with lethal alleles of wg, some defects in the wing margin occur and the alula is very reduced.
Wings about two-thirds the length and three-fourths the width of wild type, held tentlike over abdomen. Alulae absent or vestigial; proximal posterior wing margins often irregular with tendency to fold under about vein L4. Venation usually normal with occasional blistering. wgspd-fg/spd has phenotype varying from slight shortening of wings to a shape midway between the two homozygotes. Excellent viability and fertility. RK1.

wgspd-fg/wg[+] is a suppressor of visible phenotype of Scer\GAL4Bx-MS1096, hng1KK103642
wgspd-fg is a suppressor | partially of hyperplasia phenotype of ft8/ftG-rv
Sox15KG09145, wgspd-fg has abnormal size | recessive | third instar larval stage phenotype
wgspd-fg has wing hinge primordium phenotype, enhanceable by Sox15KG09145
wgspd-fg/wgspd-hL2 has wing phenotype, suppressible | partially by dshUAS.cNa/Scer\GAL4vg.PM
wgspd-fg/wg[+] is a suppressor of wing hinge phenotype of Scer\GAL4Bx-MS1096, hng1KK103642
Sox15KG09145, wgspd-fg has wing hinge primordium phenotype
One copy of wgspd-fg suppresses the wing hinge phenotype seen when hng1KK103642 is expressed under the control of Scer\GAL4Bx-MS1096. No changes in cellular proliferation are detected in these animals.
In wgspd-fg, Sox15KG09145 double mutant third instar larval wing imaginal discs, the distal hinge region is reduced in size compared to either single mutants. The rate of hinge-specific apoptosis is similar in wgspd-fg, Sox15KG09145 double mutant and Sox15KG09145 single mutant L3 wing discs.
Wing phenotype can be partially rescued by Scer\GAL4vg.PM induced expression of dshScer\UAS.cNa, rescue of the anterior wing margin and the posterior wing blade between vein 5 and the posterior margin.
wgspd-fg/wgspd-hL2 is partially rescued by wgUAS.cLa/Scer\GAL4vg.PM
Wing phenotype can be partially rescued by Scer\GAL4vg.PM induced expression of wgScer\UAS.cLa rescue of the anterior wing margin and the posterior wing blade between vein 5 and the posterior margin.

wgspd-fg behaves as a specific regulatory allele of wg in the third instar.
Regulatory mutation of wg that specifically abolishes wg function in the hinge.
Null mutations only cause etching of the alar lobe when in combination with wgspd-fg, but a mutation that affects the upstream sequences (Df(2L)DE) completely fails to complement wgspd-fg demonstrating the lesion responsible is in the upstream sequences.
