rl1/rl10 mutant third instar larvae have significantly reduced concentration of circulating hemocytes compared to wild-type controls.

rl10/rl10a heterozygotes die at the late third instar to pupal stage. Mitotic index of rl10/rl10a larval neuroblasts is clearly reduced compared to wild type. The reduction is typical of a delay in passing through interphase, as a consequence of a reduced ability to transduce proliferation signals. The ratio of metaphase to anaphase figures is elevated, suggesting the cells are being delayed at metaphase. There is a low but significant level of hypercondensed chromosomes in both anaphase and metaphase. Some anaphase defects can be seen including a low frequency of overcondensed chromatids at late anaphase, anaphase figures with lagging chromatids and randomly oriented chromatids at anaphase. In contrast to wild type, there is very little increase in the mitotic index in either rl9/rl10a or rl10/rl10a mutants after colchicine treatment for 3 hrs, and over-condensation of the chromosomes is not seen.

Clonal analysis reveals phenotypes in the adult including loss of wing vein, ectopic wing vein, reduced cell size, cell lethality and tergite bristle abnormalities.

Hemizygous larvae completely lack imaginal discs and die. Individuals die as pupae when heterozygous with rl1 or rl6.

L3 larval lethal as hemizygote no imaginal discs pupal lethal when heterozygous with rl1 or rl6 (Hilliker, 1976).

The increased concentration of circulating hemocytes characteristic for Graf1/Y mutant third instar larvae is suppressible by combination rl1/rl10, which reduces the hemocyte levels in Graf1/Y larvae to below wild-type levels, comparable to those seen in rl1/rl10 single mutants.
